Wolfporia cocos is a type of famous traditional Chinese medicine with high economicvalue, which has been widely used in Chinese traditional patent medicines andmanufacture of health food. People make use of pinewood to cuLtivate Wolfporia cocosin a long time, the cuLtivated strains are mainly based on the domestication of wildstrains. In order to make fuL1 use of the germplasm resources of Wolfporia cocos,pinewood sawdust and crop straw, esterase isozyme and ISSR molecuLar marker wereused in this research, they analyzed genetic diversity of germplasm resources incuLtivated strains and some wild strains of Wolfporia cocos. we carried on the study aboutsubstitute cuLtivation technology of Wolfporia cocos.There were 186 isoenzyme bands among 27 strains revealed by the resuLts ofesterase isozyme zymogram. The number of isoenzyme bands of each strain was 5~9bands, most strains had 6~8 bands ordinarily. There were17 zymotypes among 27 testedstrians, some strians showed the same zymotypes. The 27 stains were divided into sevengroups at certain coefficient level 81%. The 1st group: Y1 (baoshan),Yunlingl,5.78,FuLing3,A9,A10,Yuexi,T1,901,Z(z),50864,Jingzhou28,GD和GZ;the 2nd group: P0,Huazhong,E1 and FuLing 28; the 3rd group: SD; the 4th group:50478,S1,SC,50876 and DB; the 5th group: Ming006; the 6th group: L; the 7thgroup: Z (1). The similarity coefficient of some strains were very high, so it indicated thattheir genetic similarities were very close, and they may be synonym.We used 4 different original strains to select primers. We found 12 primers adaptedto ISSR. Based on the optimization of amplification system by orthogonal design andScreening of annealing temperature, 12 selected primers were used to amplify genomes of27 tested strains. A total of 85 bands were amplified, and 75.29% of which was found tobe polymorphic, with 7.1 bands per primer in average. The 27 stains were divided into sixgroups at certain coefficient level 82%. The 1st group: Y1(baoshan),A9,50478,Z(z),E1,Z(1),50864,DB,901,5.78,GZ,GD,Yunling1,FuLing3,Yuexi,Huazhong,FuLing28,Jingzhou28,A10,T1; the 2nd group: P0 and Ming006; the3rd group: SD; the 4th group: L; the 5th group: 50876 and S1; the 6th group: SC.The analysis resuLts of ISSR had proved the resuLts of esterase isozyme in certaindegree and had given some supplement to esterase isozyme on protein level. Strain L,SDand Ming006 had been divided into three independent groups by ISSR and esteraseisozyme. GZ and GD, Yunling1 and FuLing3,Z(z) and 50864, S1 and 50876 had beendivided into a group with high similarity. But the analysis resuLts of ISSR and esterase isozyme had also some differences in some extent. ISSR molecuLar markers had beenused in the study of identification and genetic diversity of germplasm resources onWolfporia cocos strains for the first time. The resuLts indicated that the technology ofISSR molecuLar marker was a simple stable high effect method, which had highpolymorphism and good repeatability. It had great application value in identification andthe analysis of relationship on the strains of Wolfporia cocos.In the study of substitute cuLtivation, the materials which included pinewood sawdustand pinewood shiver had good effect in the low and simple canopy, its survival rate was92.85%, Biological efficiency reached 18.25%. It got first primary success in thisstudy. But the yield was lowness when we cuLtivated Wolfporia cocos in shed and infield with no covers. Howere, the materials which included pinewood sawdust andpinewood shiver had better effect than cottonseed huL1 and pinewood shiver incuLtivation. We need to further research the technology of substitute cuLtivation onWolfporia cocos.
