| Objective:When the damage of microcirculation in periodontal tissue caused by periodontitis or injury,the periodontal tissue hypoxia obviously. Ischemic situation in dental lateral force has caused peiridontal tissue hypoxia at the process of orthodontic treatment.The expression of hypoxia inducible factor-1α(HIF-1α)can be activated by oxygen deficiency and some inflammatory cytokines.On the other hand, vascular endothelial growth factor(VEGF), the most important target gene of HIF-1α, plays an important role in angiogenesis and bone remodeling and repairing. The aim of this study is to study the distribution and expression of HIF-1,VEGF in normal and inflammatory gingival tissues between health adult and adult with periodontitis.Material and methods:20 Gingival tissue samples in health group from health subjects were obtained during crown lengthening surgery,20 Gingival tissue samples in test group from adult with periodontitis were collected during periodontal surgery for periodontal disease. Power VisionTM two steps immunohistochemical method with anti-HIF-1 and anti-VEGF was used to detect the distribution and cellular localization of HIF-1 and VEGF in normal and inflammatory gingival tissues between health adult and adult with periodontitis. Results:1. There was no expression of HIF-1αin the health gingival tissues in health group, while,there are positive expressions of HIF-1αin inflammatory gingival tissue of test group, difference of the expression of HIF-1αbetween two groups was statistically significant.(p<0.01).2. The expression VEGF was lower in the health gingival tissues in health group, while, it was higher and stronger in test group than that in health group, difference of the expression of VEGF between two groups was statistically significant. The expression of VEGF in epidermis of inflammatory gingival tissue from test group was significantly stronger than that in health group(p<0.01). The expression of VEGF in connective tissue of inflammatory gingival tissue from test group was statistically higher than that in health group(p<0.05).Conclusion:HIF-1 a and VEGF may participate in the periodontal tissues remodeling, and play an important role in periodontitis initiation and progression. |
