Backgroud Laryngeal squamous cell carcinoma(LSCC) is the most common malignant neoplasm in otolaryngology and increasing incidence has been observed in recent years.The pathogenesis and pathologic mechanism involved in LSCC are still unclear, neither is therapeutic response be improved significantly.With the progress in molecular biology,it is a growing recognition of the occurrence and development of LSCC and the excessive cell proliferation and reduced apoptosis.Therefore to explore the mechanism of tumor invasion and metastasis,tumor prognosis and to find the guidance to determine the clinical treatment of biological indicators has become a hot area of cancer research.Survivin is the inhibitor of apoptosis protein(IAP) family in the unique structure of a new member,is a high expression in a variety of tumor tissue specificity inhibitor of apoptosis.Survivin can regulate cell differentiation and inhibition of apoptosis in the dual function,and cell proliferative activity was positively correlated with apoptosis of tumor cells and was negatively correlated with the tumor.Survivin is also closely related to the formation of blood vessels.Nuclear factor-kappa B(NF-κB) is a multi-directional,multi-functional regulator of the nuclear transcription factor,the over-activation in tumor cells and the occurrence of anti-apoptosis plays an important role in the process.NF-κB can be induced by many apoptosis genes,including cellular inhibitor of apoptosis protein(IAP) family,Bcl-2 family members and so on. Study found that these genes inhibit apoptosis is through the combined effect of inhibiting apoptosis.NF-κB can also regulate the proliferation of tumor cells to promote tumorigenesis and development.Cyclooxygenase-2(COX-2) is the prostaglandins(PGs) synthesis of the major rate-limiting enzyme,can be arachidonic acid(AA) into prostaglandin metabolism product of the body involved in the pathophysiological process of multiple.Study shows that,COX-2 play an important role in tumor occurrence,development and metastasis.COX-2 in tumor happened in the high expression can adjust metabolism of carcinogens,apoptosis,immune surveillance,angiogenesis,such as a number of ways to promote the occurrence and development of tumor.Study found that Survivin,NF-κB and COX-2 in inhibiting the three apoptosis and cell cycle regulation plays an important role,and in the promotion between tumor growth,proliferation and metastasis may have a certain degree of mutual relations.So far,the LSCC has been reported in fresh Survivin,NF-κB and COX-2 relationship between the literature.ObjectiveTo discuss Survivin and NF-κB,COX-2 gene expression in laryngeal squamous cell carcinoma of the relevance,further defined the occurrence of laryngeal squamous cell carcinoma invasion and metastasis and molecular mechanisms,with a view to laryngeal squamous cell carcinoma of the gene targeted therapy to provide theory.MethodsThe expressions of Survivin,NF-κB and COX-2 in tissues of 50 cases,7 cases of vocal cord polyp(VCP),5 cases of margins of LSCC were studied by Immunohisto-chemical technique(S-P) to analyze their correlations with the occurrence and development of LSCC.SPSS13.0 software was employed to analyze all data.Statistical evaluation was performed using Chi-Square test.P<0.05 was considered as statistical significance. Results1.The expression of Survivin protein was localized in the cytoplasm.The positive expression rates of Survivin in LSCC tissues of 50 cases,7 cases of vocal cord polyp(VCP),margins of LSCC were 56.0%(28/50),0(0/7) and 0(0/5) respectively. Comparing the staining rate among those 3 groups,there were significant difference.The positive rates of LSCC was significantly higher than those of 2 groups(P<0.05);The expression of Survivin had significant relation with lymphatic metastasis,pathological grading and TNM staging(P<0.05).2.The expression of NF-κB protein was localized in the cytoplasm.The positive expression rates of NF-κB in LSCC tissues of 50 cases,7 cases of vocal cord polyp(VCP),margins of LSCC were 46.0%(23/50),0(0/50) and 0(0/50) respectively.Comparing the staining rate among those 3 groups,there were significant difference.The positive expression rates of LSCC were significantly higher than those of 2groups(P<0.05);The expression of NF-κB had significant relation with lymphatic metastasis and TNM staging(P<0.05),but had nothing to do with the pathological grading(P>0.05).3.The expression of COX-2 protein was localized in the cytoplasm.The positive expression rates of COX-2 in LSCC tissues of 50 cases,7 cases of vocal cord polyp(VCP),margins of LSCC were 42.0%(21/50),14.3%(1/50),0(0/50) respectively.Comparing the staining rate among those 3 groups,there were significant difference.The positive expression rates of LSCC were significantly higher than those of 2 groups(P<0.05);The expression of NF-KB had significant relations with lymphatic metastasis and TNM staging(P<0.05),but had nothing to do with the pathological grading(P>0.05 ).4.There were significantly positive correlations among the expression of Survivin,NF-κB and COX-2 in laryngeal squamous cell carcinoma tissues(P<0.05).Conclusions1.The high expression of Survivin,NF-κB and COX-2 in laryngeal squamous cell Carcinoma tissues may promote the growth,proliferation and metastasis of laryngeal squamous cell carcinoma.2.The expression of Survivin protein had positive correlation with that of NF-κB and COX-2 protein in LSCC.United detection of the expressions of Survivin, NF-κB and COX-2 contributes to the diagnosis,therapeutics and prognostic assessment of laryngeal squamous cell carcinoma.
