The protective effects of chloroform extracts from Isatis indigotica leaves on the damage of embryo development and male germ cells induced by cyclophosphamide (CP) were studied in mice. Methods: The pregnant mice weight, the fetus weight and the fetus body and tail length were measured, the survival, death and absorb rates of fetus were calculated by embryonic teratogenicity tests. The fetus skeletal development and visceral abnormality were observed by skeletal staining and visceral section .The rates of spermatozoon malformation and micronucleus of early spermatids were detected by spermatozoon malformation and micronucleus tests. Results: Isatis indigotica itself has no embryotoxicity and genotoxic effects, but has significantly protective effects to embryonic development and germ cell damage induced by CP. The group of chloroform extracts at the dose of 100mg/kg.w+CP10mg/kg.w could obviously increase the survival rate (increase 59.60%), decrease absorb (decrease 86.91%), death and malformed rate . Further more, it could also increase the fetus's body and tail length as well as fetus weight .The inhibitory rate of spermatid micronucleus and spermatozoon malformation is 57.38% and 50.87% respectively. Conclusion: The Chinese medicine Isatis indigotica has the protective effects on embryonic development and inhibitory effects on germ cell damage in mice.
