Objective This study was designed to investigate the protective effects of Ipomoea Batatas Poir Cv anthocyanins against 60Co ? ray-induced irradiation damage in mouse thymocytes. Methods We first established an irradiation damage model of mouse thymocytes by using 60Co ?-ray (3650uJ/cm2). The morphologic changes of thymocytes were investigated by using transmission electron microscope and the DNA ladders were determined by agar gel electrophoresis. The concentrations of GSH-Px, SOD, MDA, T-AOC and cellular viability were determined by biochemical methods. The free calcium, mitochondria memberine potential and the rate of apoptosis on thymocytes were tested by using flow cytometer (FCM). Moreover, the expressions of p53, Bax and Bcl-2 proteins were examined by immunocytochemical technica. With the technique of in situ hybridiation, we also tested the expressions of p21, p38 and mRNA. Furthermore, the scavenging properties of Ipomoea Batatas Poir Cv anthocyanins on superoxide anions(O2-), hydroxyl radical(OH ? ), peroxynitrite (ONOO-) and free radicals of lipids were observed by using chemiluminescence and electron spin resonance(ESR) techniques. Results Ipomoea Batatas Poir Cv anthocyanins 0.625 2.500g/L could obviously enhance the activities of GSH-Px, SOD, A-SAC, T-AOC . The concentration of cellular free calcium, ROS and MDA and the rate of apoptosis was significantly decreased by Ipomoea Batatas Poir Cv anthocyanins. In addition, Ipomoea Batatas Poir Cv anthocyanins (0.625 - 2.500g/L) distinctly raised the expression of Bcl-2 gene and decreased the expression of P53, Bax. And mRNA expressions of p21 and p38. The ultrastructural damages of the cells were also markedly inhibited. Results showed that Ipomoea Batatas Poir Cv anthocyanins could effectively scavenge O2- (IC50=1.2 x 10-6g/ml X OH ? (IC50=4.48 x 10-7g/ml), ONOO- (IC50=1.0 x 10-6g/ml) and free radicals of lipids (IC50=4.4 x 10-5g /ml) (P<0.05). Conclusion Ipomoea Batatas Poir Cv anthocyanins has protective effects against damages of thymocytes caused by irradiation of 60Co?-ray. Its protective effects might be related to its inhibition of ROS and the lipid peroxide , the raising expression of Bcl-2 gene, the decreasing expression of p53 and p21 gene, the increasing anti-oxidative enzymesin cells and so on.
