With the over-exploration of wild Radix Scutellariae, the source of which was serious destroy-ed and cultivated Radix Scutellariae became the main commodity. Cultivated species is not selected in the long-term production, resulting in the aggregate yields and quality of medicine uneven and the quantity of chemical constituents instability. Therefore, it is necessary to carry on the species screening work. It were made and found out that a comprehensive analysis of quality of different districts Radix Scutellariae and the relationship between gene and quality. The genetic characteristic-cs of Radix Scutellariae from the molecular level were revealed, and to provide some references for selecting high-quality, high-yielding species of Radix Scutellariae in the future.In order to select good quality seed sources of Radix Scutellariae, wild and cultivated Radix Scutellariae from seven districts were collected. A comprehensive comparison of several compounds was made, including determining the content of total flavonids and polysaccharide by UV spectru-m, the content of baicalin, baicalein, wogonin by high performance liquid chromatograph (HPLC) in the collected Radix Scutellariae, establishing HPLC fingerprint of the flavonid in Radix Scutellar-iae, observing the microscopic structure of Radix Scutellariae from different districts by paraffin s-ections, detecting the genetic distinction among Radix Scutellariae from different districts by ISSR.The DNA fingerprint library of different districts Radix Scutellariae were established. The relations-hip between the genetic characteristics and the quality were studied.Results and discussions:(1) Through determining the contents of the baicalin, baicalein, wogonin, total flavonids and polys-accharide in Radix Scutellariae which were collected from different areas, it was initially evaluated 4 fine samples on the basis of the evaluation of chemical quality by gray pattern recognition me-thod, including cultivation from Hebei kuancheng; cultivated two-year-old from Shanxi fenyang; wi-ld from Shanxi fenyang and wild from Shanxi lishi.(2) Through observing cross-section of the microscope and appearance characteristics of the sampl-es , the results is as follows: the 4 fine samples which were elected on the basis of the evaluate-on of chemical quality accord with traditional evaluation that it is excellent by long, solid quality and yellow samples.(3) Through establishing the HPLC fingerprint, the results are as follows:①The peak areas are hi-ger of No. 1,2,10(luteolin),15(wogonoside),17,18,19(baicalein),21(wogonin),22(chrysin),23(skullcapflavoneⅡ),24(oroxylin A) in the 4 fine samples which had been selected.②There a-re 2 stable peaks (No.3a, 3 peaks) in wild Radix Scutellariae from Chengde,but the other samples is only one peak (No. 3 peak) in this location. It is preliminary pointed out that Chengde wild R-adix Scutellariae sample's fingerprint characteristic is emerged a stable chromatographic peak at 15.7min (No. 3a).(4) It is used ISSR(Inter-sample Sequence Repeat, ISSR) molecular marker to analyse the genet-ic relationships of samples from different areas. The 4 fine samples that is preliminary selected ga-thered in a category, and the samples which were gathered with them are of better chemical qual-ity. It is pointed out that there are relevant between chemical quality and gene. It is similar in ge-ne characteristic of samples of which chemical quality better, and there exist some differences bet-ween those and the samples of which chemical quality worse. It is provided the gene level suppo- rt in selecting excellent varieties of Radix Scutellariae.
