Rhizome Belamcandae is the dried rhizome of a medical plant Belamcandachinensis (L.) DC. from Belamcanda genus, iridaceae. It is a commonly traditionalChinese herbal medicine, and a genuine drug in hubei. It used to treat flu, cough,inflammation, asthma, pneumonia as well as throat troubles such as tonsillitis,pharyngolaryngitis, laryngeal edema and so on.Reports and documents studied on various researching field in past few yearsabout Belamcanda chinensis (L.) DC. Were summarized. We studied on idioplasmresources of Belamcanda chinensis (L.) DC. in following aspects:①Studied on thegenetic diversity of Belamcanda chinensis (L.) DC. which from different regions byusing the method of RAPD.②Constitute the Preserved standard for BlackberrylilyRhizome of Belamcanda chinensis (L.) DC. And the results obtained as follows:1. In the experiment, five essential factors which may affect the result ofRAPD were optimized by orthogonal experiment and three single factor experimentsof annealing temperature, annealing times and cycle times. The result showed that theoptimal conditions of RAPD-PCR system.2. There were 100 random primers (10-mers) used for the PCR amplification ofgenomic DNA fragment. Fourteen of the primers could amplify polymorphic bands:96 of 126 amplified bands were polymorphic, accounts for 72.6%. Every primercould amplify 4 to 9 polymorphic bands, with an average of 6.9 bands. This indicatedthat the genetic diversity of Belamcanda chinensis (L.) DC. from different regions ishigh.3. The value of the similarity matrix of 16 accessions were 0.073 to 1.000. Itshowed that the larger its number value is, the nearer the hereditary distance could be.Cluster analysis showed that the samples could be classified into 5 clusters. The resultindicated that the smaller the geographic distance between two Belamcanda chinensis(L.) DC. samples, the smaller their genetic difference, while the genetic differencebecame greater as their geographic distance increased. However, it should not beignored that the natural environments, especially the local ecological conditions mayalso show some influence on the genetic variations among different Belamcanda chinensis (L.) DC. samples. It is obvious that the result of cluster analysis based onRAPD of Belamcanda chinensis (L.) DC. are more coincide with geographicaldistribution of Hubei Province.4. Through the consult researches, the information collection, investigate andreorganizes this project findings, constitute the Descriptors and Preserved standardsfor Blackberrylily Rhizome of Belamcanda chinensis (L.) DC., also regulate theenvironment of cultivation, suitable condition, technology and method in cultivation.
