Pinellia ternata(Thunb.)Breit which is a main material of processed Chinese traditional medicine has been applied for a long time. As a precious wild medical resource with low self-propagation coefficient, the supply quality of Pinellia ternata (Thunb.)Breit can't meet the growthing demands in the domestic and export market at present. Though human has cultivated Pinellia ternata(Thunb.)Breit for at least 30 years, the cultivation of it is still seriously affected by circumstance, disease and cost, which caused difference on the quality and the succession genetic message that need to be researched. Hubei Province is the one of main producing regions for Pinellia ternata(Thunb.)Breit, which not only has abundant wild resource but also has the special variety that own premium quality and yield. The question is: How about the variability between the wild plants and cultivated ones? Whether this variability will cause the change of species and the losing of genetic message? Obviously, all of such questions are blank in relative research at present.For gaining the answer, we collected 19 samples from 8 regions in Hubei Province and 2 samples from Lushan in Jiangxi Province to analyze the genetic diversity. Furthermore, we collected 6 samples from Enshi regions which have high yield to research the relationship among them. Our work is significant to cultivation and quality identification in theoretics and practice.Improved CTAB method was used to extract DNA from fresh leaves. Many parameters of RAPD technique was studied by a series of experiments, including DNA extraction, optimization PCR amplification by comparing a series of annealing temperature, template concentrations, amounts of Taq DNA polymerase and so on, selection of ten from eighty random primers. We studied on these resources in following aspects: ①studied on the external characters and cultivation of Pinellia ternata(Thunb.)Breit. ② studied on the genetic diversity of Pinellia tanata (Thunb.)Breit which from different regions by using the method of molecular biology. ③studied on the relationship of Pinellia ternata(Thunb.)Breit from Enshi region. And the results obtained as follows:1. Random Amplified Polymorphic DNA (RAPD) was used to research genetic diversity and relationship among the samples from different regions. And 67 sites were detected while 59 were polymorph, which account for 88.06%. This indicated that the genetic diversity of Pinellia ternata(Thunb.)Breit from different regions is high.2.We had the rescaled distance cluster combine and the proximity matrix, it showed that the relationship among the samples from different regions of Pinellia ternata(Thunb.)Breit. The main factor which affects the genetic distance is the different habitat rather than different cultivation pattern.3. The samples from Enshi region can be gathered into same cluster although they have different modality of leaves, which indicated different localities have more influence on genetic diversity than different modality of plants.
