| Objective:While the fast aging of population,prevalence rate and the effection on society and family of Alzheimer's disease(AD)are cincerned widely.Theregore, deeply studying on patnogeny and mechainsm of AD have much more clinical worthy and social significance,for exploring effection therapy measure and medication.So we investigate the therapeutic mechanism of Qiancengta Decoction deeply,to provide traditional chinese medicine and national medicine with experimental accordance and clinical guidance.Methods:Male,Wistar,13-month Rats(n=60)were randomly divided into six group.That is normal group,blank group,AD model group,piracetam treated group, high-dose of Qiancengta Decoction(HQD for short)and low-dose of Qiancengta Decoction(LQD for short),10 rats for each group.After feeding in ordinary ways for one week,except the normal group,AD model group,piracetam treated group, HQD group and LQDgroup(the four group)received intraperitioneral injection of D-galaetose(100mg/kg.d)for six months,and the blank group rats with lavage.At the beginning of the seventh week,the four group rats were injected Amyloid-β25-35in the bilateral Hippocampus.The blank group was injected the same dose of lavage.After 24 hours,every group was treated with different madicine for four weeks.And then,Morris water maze(MWM)was used to observe the ability of space leaming memory.By the light microscopy and electron microscopy to observe the changes of morphology and structure in the hippocampus.By the method of immunohistochemistry detect the expression of C-FOS protein.Results:In the behavior experiment,not all the ability of leaming gain and the competence of information reserve,the significant differences were observed between the model group and normal group(P<0.01),also between every treated group and model group(P<0.01),and also between the HQD,LQD group and piracctam treated group(P<0.01).These indicate that the AD model in this experiment is successful and Qiancengta Decoction can improve the AD rats' ability of learning gain and the competence of information reserve more efficiently. By the light microscopy and electrical microscopy observation,sum of neuron of model group was less than that of normal group.programmed cell death could be seen clearly,the neurocyte decreased and deformed in model group rats.Qiancengta Deeoetion can ameliorate this pathological changes.Detected by immunohistochemistry,the contents of C-FOS protein in AD model group rats obviously increased compared with normal group rats(P<0.01).Every therapy group could decrease the contents of C-FOS protein(P<0.01),but these alterations were more obversly with High-dose of Qiancengta Decoction treatment(P<0.01).Conlusions:Kidney-essence and marrow weak,phlegm and blood stasis is the basic etiological factor for AD,and replenishing kidney-essenee and marrow, removing phlegm and promoting blood circulation therapy is operative on treating AD. Qiancengta Decoction has the therapeutic mechanism,so it can treat AD efficiently.And its mechanism could represent that increased the learning memory-level,better the pathology change,decreased the expression of C-FOS. |
