Objective:1.To set up the CFS model of rats by chronic multiple factors stress.2.To investigate the correlativity between the chronic multiple factors stress and the HPA axis function of the CFS model of rats.3.To discuss the possible mechanism of acupuncture in effect on the HPA axis.4.To explain the pathogenesis of CFS and the regulatory mechanism of acupuncture from the aspects of psychology and HPA axis.Methods:Totally 50 SD healthy female rats.The double factors and multiple factors with chronic stress were used to set up the CFS models respectively.After the establishment,the models of two groups were compared.The rats in the acupuncture group were acupunctured in bilateral Baihui,Zusanli,Taichong alternatively,and were given the tress factors at the same time.The behavior changes of the rats were observed.The content of the serum CORT in was detected by radioimmunoassay and the content of serum ACTH by enzyme-linked immunosorbent assay.The content of the CRHmRNA in hypothalamus was detected by polymerae chain reaction.Results:1.Decreases of weight,body temperature;the activities of rats in open-filed test;and shortening of exhausting swimming time,as well as the reduction of contents of the CORT and ACTH in serum in the multiple factors model group were more obvious,which showed that the models were closer to clinical manifestations of patients with CFS.2.The weight,body temperature,the activities of rats in open-filed test,and exhausting swimming time of rats in acupuncture group were closer to the normal control group,which indicated that the acupuncture therapy had better effect than the model group in treating the physical fatigue and the mental fatigue.3.After acupuncture treating,the contents of the CORT and ACTH in serum,the CRHmRNA in hypothalamus of the rats in the acupuncture group were much lower than those in the model group,lt proved that the acupuncture therapy can markedly regulate the neuroendocrine system,reduce the excitability of HPA axis.Conclusions:1.The behavior and the indexes from HPA axis change of the multiple factors model of rats with CFS is closer to clinical manifestations of patients with CFS.It can be used as an animal model to the CFS research.2.The study proved that the hyperaction of HPA axis,that lead to the dysfunction of negative feedback mechanism is may be one of the pathogenesis of CFS.3.The method of acupuncturing Baihui,Zusanli,Taichong can improve the fatigue states of the rats,markedly reverse the hyperactive HPA axis.It is an effective therapy of CFS.
