Objective:To investigate the effects of different exercise loads on the synthesis mechanism of testos terone in male rats, and to reveal some intervention mechanism of aerobic exercise on male rat s in vivo androgen, reveal the effect of aerobic exercise on rat endocrine, in order to reduce t he athletes exercise-induced low serum testosterone phenomenon, improve theathletes sports abil ity, enhance the muscle strength, provide theory supports the elimination of sports fatigue. Methods:Totally 37 healthy male Sprague-Dawley (SD) rats were randomly divided into 4 groups: normal control group (C, n=8), the general exercise group (CE, n= 10), long time exercise grou p (LE, n=10), each group of unified common feed. Motion scheme for adaptive swimmingfor 1 weeks, C group with no training, group CE 60min, group LE 120min swimming exercise,6 days a week, a day of rest. Detection of total cholesterol in serum of rats after the end of the ex periment (TC) and determination of serum testosterone ELISA method (T) and corticosterone (C ORT), leptin (LEP), GnRH, LH, FSH content, determination of tissuecholesterol ELISA metho d (CH), testosterone (T), P450scc and stAR expression by P450sccand stAR; detection of RT-P CR in rat testis tissue volume; observation of the change oftesticular tissue morphology and H E staining.Results:l.No significant differences in body weight before the 1 training rats after 10 weeks of traini ng,two training group rats compared with the control group showed significant difference. After 10 weeks, compared with the C group CE testis coefficient has extremely significant differen ce,and there was significant difference between the LE group.2.Testis tissue in C group of seminiferous tubule development basically normal, spermatogeni c cells arranged very closely, the structure is clearly visible, spermatogonial cellsclose to the epi thelial basement membrane, the proliferation of normal, all within the lumen ofspermatogoniu m, primary spermatocyte, spermatid stages of cells can be clearly seen. While the CE group co mpared with C group, the testis seminiferous tubules and sperm andspermatogenic cells, the arr angement is more loose, LE group than in CE group was more. In the LE group also found th e cell falls off phenomenon, although this phenomenon is less, but this phenomenon in the cont rol group is more scarce. But in group LE, the normal connectionwith the basement membrane of the spermatogenic cell separation, occurred off. Normal control group of Leying cell cytopla sm was abundant; while in CE group, called the control group cells were less in group LE les s.3.10 weeks of training, no difference in the serum total cholesterol, testis cholesterol and Cg roup and CE group than in the LE group was significantly lower. Compared with C group,C E group, T in serum of rats decreased slightly but no significant difference in LE group, the dif ference was extremely significant; compared with CE group and LE group also hadsignifican t. Compared with the C group in testis of T, the LE group had significant difference.4.The serum corticosterone (CORT), leptin (LEP), GnRH, LH, FSH compared with the control group had significant changes, corticosterone (CORT), leptin (LEP) increased, while GnRH,LH, F SH decrease phenomenon.5.And the content of Cin the testis of testosterone synthesiskey enzyme of stAR group LE group significantly decreased, compared with the CE group, significant decrease. The content of P450scc and C group and CE group was significantly lower than. Testosterone synthesiskey enzyme of mRNA expression of stARmRNA, P450sccmRNA and C group compared with C E group and LE group were significantly different.Conclusion:1.Long time high load exercise may cause morphological changes intesticular tissue, thereby af fecting the secretion of testosterone.2.Effects oflong time high load exercise secretion, which may CORT、LEP through HPG axis a pproach or directly into the testis tissue regulation, influence of testicular testosterone secretion.3 High load exercise may affect the effects of testosterone synthesis keyenzyme mRNA levels of enzyme expression, and then the interferencetestis testosterone synthesis.
