Effects Of Six-week Aerobic Exercise On Hepatic Leptin Receptor In Obese Rats

Objective: This study involved six-week aerobic exercise combined with dieting to intervene in high fat diet-induced female(SD)obese rats to observe 6-week aerobic exercise and dieting on obese rat body indicators and viscera.The effects of fat and serum leptin concentration and liver leptin receptor expression were analysed by enzyme-linked immunosorbent assay and immunohistochemistry.To explore the effects of exercise and diet on serum leptin and hepatic leptin receptors in obese rats under high-fat diet,and to reveal the changes of hepatic leptin receptors in rats with aerobic exercise and diet on high-fat diet.To provide a theoretical basis for the development of a more reasonable exercise weight loss program and treatment of liver metabolic diseases.Subjects and methods: 130 SD rats(200~220g)were purchased from Beijing Weitong Lihua Biotechnology Co.,Ltd.,and they were divided into normal control group(NC,N=8),fed ordinary feed;obesity module(OB,N = 132),fed a high fat diet for 8 weeks of obesity modeling.At the end of modeling,48 rats that met the obesity criteria were randomly divided into obese control group(OC),diet group(D),exercise dieting immediate group(ED0),exercise dieting 3h group(ED3),and exercise immediate group(E0),exercise 3h group(E3),each group had 8 rats.Except for the NC,the other groups were fed with high fat diet.A 6-week swimming dietary intervention was performed.Rats in the exercise group were fasted for 3 hours immediately after exercise and after exercise for 3 hours.Serum leptin and hepatic leptin receptor were determined by enzyme-linked immunosorbent assay,and the liver leptin receptor-positive cells were semi-quantitatively analyzed by immunohistochemistry.All data were statistically analyzed with SPSS21.0.± Standard deviation(?X ± S)indicates that P < 0.05 is a significant difference.Results:(1)After the end of the 8-week modeling,obese rats meeting the obesity criteria were successfully screened by combining Lee's index and body weight.(2)After 6 weeks of experimental intervention,the body weight,Lee's index and perirenal fat of group D and ED group were lower than those of OC group and NC group;body weight,Lee's index and perirenal fat of group E were lower than OC group but high.In the NC group.(3)After 6 weeks of experimental intervention,serum leptin were significantly higher in group D,ED0 and E3 than in the NC group(P<0.01).Serum leptin was significantly higher in ED0 and E3 groups than in the OC group(P< 0.05);OC group,ED3 group and E0 group were higher than OC group,but there was no significant difference.The ED3 group was lower than the ED0 group,but there was no difference;the E3 group was higher than the E0 group,and there was a significant difference(P<0.05).(4)After 6 weeks of experimental intervention,the liver leptin in the OC group was lower than the other groups,the ED0 group was higher than the other groups,the ED3 group was lower than the ED0 group,and the E0 group and the E3 group were lower than the D group,the ED0 group,and the ED3 group.There were no meaningful differences between the groups and the above groups.Conclusion:(1)Long-term high-fat diet causes obesity in rats and a significant increase in body fat mass.Aerobic exercise and diet can minimize the body weight and Lee's index of obese rats,and reduce visceral fat content.The weight loss effect is good.6 weeks of simple aerobic exercise can effectively control the growth of high fat diet,but it can not achieve weight loss..(2)High-fat diet will increase serum leptin concentration,exercise and diet can also increase serum leptin concentration,serum leptin concentration will decrease after exercise for 3h,and serum leptin concentration will increase after 3 hours of exercise alone.It shows that the combination of simple exercise and exercise diet will have different effects on the changes of serum leptin,which has a certain relationship with exercise intensity and body energy regulation after exercise.(3)The high-fat diet did not significantly alter the content of hepatic leptin receptors.Although exercise and dieting had a certain effect on increasing the expression of leptin receptor in the liver,there was no significant difference compared with the control group,related to exercise and diet.Further research is needed on the intervention approach.
