Objective To observe the effects of the Ginkgo Biloba Exocarp ingredients [Polysaccharides (GBEP) , Ginkgolides and Ginkgolic acid] on the physical performance of the aging mice induced by D-galactose and to research into their mechanisms of deferring aging and improving moving ability.Methods The assay methods for antistatic fatigue and loading-weight exhausting swimming were used for the study of physical performance ability. The learning and memory ability was measured by using climbing-lever test and jumping-stand test. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in Hippocampus, the activity of glutathione peroxidase (GSH-Px) and acetylcholinesterase (AChE) in cerebral cortex were measured. The viability of PC12 culture cell was monitored by MTT assays.Results GBEP and Ginkgolides can ameliorate the physical performance of aging mice in antistatic fatigue test (P<0.05) and loading-weight exhausting swimming test (P<0.05), improve the ability of learning and memory in climbing-lever test and jumping-stand test (P<0.05). At the same time, the activity of SOD and GSH-Px were increased (P<0.05), the level of MDA and AChE were reduced (P<0.05) respectively. The effect of Ginkgolic acid on the aging mice was not obvious.Treatment of PC12 cells with hydrogen peroxide caused a great decrease in the A490 value. GBEP and Ginkgolides dose-dependently improved the A490 value (P< 0.05), respectively. GBEP in combine with cAMPcould increase the viability of PC12 cells (P<0.05), but there is no great effect with verapamil. Ginkgolides in combine with cAMP enhanced the viability of cells (P<0.05). But with verapamil, the enhancement of Ginkgolides on the viability of cells was inhibited.Conclusions 1. GBEP and Ginkgolides can ameliorate the physical performance of aging signs induced by D-galactose and enhanced the ability of learning and memory inthe aging process.2. GBEP and Ginkgolides can rainforce the antioxidant ability , improve the symptoms of oxidative damage and the cholinergic function in the brain of aging mice.3. The effect of GBEP possibly was carried out through the cAMP signal transduction path. The effect of Ginkgolides concerned with both signal transduction paths of the cAMP and Ca2+.
