Endometritis is a postnatal reproductive disorder disease in cows. Postpartum uterus suffer bacteria pollution including conditions pathogenic bacteria, whether cause endometritis or not, depends on the uterus of immune state. Mast cells (MC) regulates function of uterus, its number and granule state change regularly as uterine reproductive cycle, and effect structure and function of endometrium. Therefore, structure of endometrium what counts is to uterine immunity are an important factor in endometritis happerned. MC play biological functions through its mediator, and participate in inflammatory reactions.It was reported that histamine was detected in urine as diagnose evidence of endometritis. At present, researchs focus on the role of MC in infection and uterine physiology. However, the role of MC mediator in endometritis was not known.For this reason, We selected suffering from acute endometritis cows, The LTB4, HA, IgE,PGF2α, P4 and E2 concentration was detected by ELISA, H1R, H2R, ESR-1, PGR and BLT2 mRNA expression were examined using real-time PCR in the bovine endometrium, and the endometrial ultrastructure was checked by transmission electron microscopy. We could investigate the action of MC mediator in endometritis, for revealing pathogeny of endometritis lay the foundation. The results showed as follows:1. In endometritis cows PGF2αconcentration were 9.949±0.831 ng/mL, significantly lower than 40.525±10.847 ng/mL (P<0.01), P4 concentration were 17.108±0.507 ng/mL, control groups were 8.734±0.542 ng/mL, the endometritis groups were significantly higher than control groups (P<0.01), shows that PGF2αwhich was derived from uterus reduced, inhibited corpus luteum subsidise, lead to P4 concentration was higher.2. In endometritis cows the concentration of LTB4 were 78.842±12.552 pg/mL, HA were 74.378±3.962 ng/mL, the control groups were respectively 23.143±1.117 pg/mL, 32.547±4.054 ng/mL, they were extremely significant between-group differences(P<0.01). The concentration of IgE were 11.398±1.342μg/mL in endometritis groups, control groups were 6.596±0.202μg/mL, the endometritis groups were significantly higher than control group(P<0.05), shows that high concentration of LTB4, HA in endometritis involving IgE.3. In the electron microscope, suffering from cows endometrial MC Nuclei become pyknotic, degranulation and vacuole. In control group MC nucleolus darity, granule were homogeneous distribution. This result shows that endometritis endometrium MC release media, participation in the uterus of local defense. 4. Endometritis cows suffering from H1R mRNA expression level significantly lower than control groups(P<0.05); but the endometritis group H2R mRNA expression ratio was significantly higher than the control groups(P<0.01); meanwhile, BLT2 mRNA was significantly higher than control groups(P<0.01). Shows that endometrial H1R and H2R mRNA expression imbalance to reduce the level of uterine immunity; however, LTB4 caused by BLT2 endometrial pathological damage.5. Endometritis cows suffering from ESR-1 mRNA expression level significantly lower than control groups(P<0.01); but the endometritis group PGR mRNA expression level was significantly higher than control groups(P<0.01); shows that PGR and ESR-1 could inhibit the uterus to contract. In conclusion: IgE induces endometrial MC degranulation in endometritis, make LTB4 and HA concentration significantly increase, it maybe exist Type I allergic reactions. Endometrial H1R and H2R mRNA expression imbalance to reduce the level of uterine immunity, BLT2 mRNA high expression results in endometrial pathological damage. PGR mRNA higher expression and ESR-1 mRNA lower could inhibit uterine contractions, it maybe concern with PGF2αreduced. While, how high concentration P4 affect MC PGR in endometritis, and regulate MC release medium should be further researched. Therefore, endometrial MC by its receptor effect physiological function of endometrium, further plays important role in the development of endometritis.
