| Objective:Malignant tumor has been seriously threatening human being's health and life,it inevitably to lead to death without control.Now the treatment to tumor is the synthetic application of surgery,radiotherapy,chemotherapy and biotherapy and so on,but all treatments have their own indications and contraindication.Although some patients on early stages have a chance to completly cure,but because of some kinds of reasons,many patients have been belonged to advanced stage when discover,they have been missed the best time for treatment.The chemotherapy have certain efficiency and has an significant position in the treatment of the patients with advanced tumor.In practice,they also have some problems for their severe negative effects,high price and multidrug resistance,these can lead to the aggravation of patients' conditions and the degression of quality of life,even lead the patients to give up.It is well known the development of all tumor is a anvanced,very long process.We hope can obtain the opportunity to cure cancer or enhance the immune function of organism maximatily,stable and control tumor,alleviated the harm of tumor,make the patients keep a better quality of long-range,the existence of band-tumor for long-term has been our reality. So,we have tried our best to research one convenient feasible medicines or methods,which not only to prevent or alleviated the growth of tumor,and also safely,can long-term application in following treatment.Traditional Chinese medicines is greatful and precious,have their merits such as widespread medicinal herbs resource,little toxicity and low price etc.In recent years,traditional Chinese medicines treat tumors in a systemic way has been accepted by more and more experts and many patients,number of scholars not only domestic but also abroad were interested in studying Chinese medicine for the treatment of malignant tumors. Chinese herbal medicines have achieved encouraging results,the role of anti-tumor medicine from a single drug to compound preparation,which is effective in the combined therapy of malignant tumor.Yiliuning is a traditional Chinese compound prescription composed of Rab,Aca,Duc and Sol,which is a clinical experienced prescripsion of our technical office.It has definite affirmative effect on the treatment of tumours for digestive system,and has little toxicity.According to our former experimental in vitro anti-tumor and result of its four compositions' extracts which compound with Rab,Aca,Duc and Sol can control growth and multiplication of tumor cells;induce tumor cells to apoptosis and arrest the Eca109 cells during S-phase,the mouse's acute toxicity experiments have shown when maximatily dosage is 126 times than clinical,Yiliuning have no obviously toxicity in short time,and the pharmacodynamics experiment has confirmed that tumor inhibition rate of Yiliuning were greater than 30% to H-22 and S-180.The objectives of experiment are to observe the effect of Yiliuning's antitumor and potential mechanism of action on human esophageal cancer EC9706 bearing nude mice, provide experiment basis and evidence for the use of clinic.Methods:The experiment of antitumor on EC9706 bearing nude mice and immunohisto-chemistry.The model of nude mice bearing tumor were established by human esophageal cancer EC9706 cells.Tumor bearing nude mice were divided into five groups with randomly and each group consisted of eight nude mice:Negative control group(the same volume of physiological saline was affused by mouth);Positive control group (through peritoneal injection PYM 4mg/kg, every other day);low,moderate,high three dosage group of Yiliuning(7.47g/kg.d,14.94g/kg.d,22.41g/kg.d extracts of Yiliuning were respectively affused by mouth).After giving drugs for 28 days (four weeks) continuously,from first day observe the change of nude mice weigh,tumor and some commonly things.29d mice were sacrificed with cervical dislocation,we took the tumor tissues to weigh,computed the rate of inhibiting tumor.At the same time make paraffin block pathological section,use the immunohistochemistry with SP staining method to detect every group's vascular endothelial growth factor (VEGF) and endostatin (ENS) expression.Took heart,liver,spleen,lung,kidney,spermary through HE dye to observe the change of primary organ and tectology with the optical microscope,understand the toxic performance of drugs to organs.Results:The tumor inhibition ratios of positive control group and Yiliuning respectivly group with low,moderate,high dosage are 47.40%,35.81%,36.72%,42.3%.Compared with negative control group,difference of all groups are statistically significant:the groups of positive control group and Yiliuning with low,moderate,high dosage (P< 0.01).The Yiliuning groups respectivly have no dose-effect relationship.When the experiment is end,the negativ group and Yiliuning group's weight all higher than the e positive control group.The results of organ and tectology indicate that there are Yiliuning with high dosage have no obviously toxic effect to main organs of mouse.The expression frequency of VEGF of Yiliuning groups and positive control groups are lower than negative control group.Compared with the negative control group,the difference of the Yiliuning groups and positive group are statistically significant (P<0.01).The expression frequency of ENS of positive control groups and Yiliuning groups are high than negative control group.Compared with the negative control group,the difference of the Yiliuning groups and positive group are statistically significant (P<0.05).The Yiliuning groups respectivly have no statistica-lly significant (P>0.05).Conclusion:1. Yiliuning can improve body weight and general state of the nude mice bearing EC9706.2. Yiliuning can obviously resist the growth of tumor of the nude mice bearing EC9706.3. The mechnism of anti-tumor may be control the expression of VEGF protein and endostatin to inhibition of tumor angiogenesis so as to achieve the growth of antitumor effect. |
