Objective:To establish the cultured modle of microencapsulated human gastric carcinoma (HGC) cell lines SGC-7901, and investigate the effect of microencapsulation environment growth features and gene expression of HGC cells.Methods:The HGC cell line SGC-7901 was cultured and encapsulated in APA microcapsules using the prepared electrostatic droplet generator. The level of proliferation and metabolism of the microencapsulated HGC cells were detected using MTT and glucose-lactic acid measure, respectively. Microencapsulated HGC cells were stained with HE and expressions of BrdU, VEGF and PCNA gene were detected by immun-ocytochemistry on the 7th day, the 14th and the 21st day, monolayer cultured cells as control.Results:The microencapsulated HGC cells showed clumpy during 48-72 h. On the 14th day, the celluar mass diameter reached 300μm under inverted microscope; At the same time, the relative number of the cells were no longer increased and declined slightly on the next few days by MTT. Glucose was consumed and lactic acid was generated by microencapsulated HGC cells with the cultured time increased. On the 20th days, the lactic acid content was decreased at 10mmol/L and the glucose content was increased at 100mmol/L by detecting the supernatant of culture medium. The expression of PCNA and VEGF gene could be detected in SGC-7901 cells cultivated with monolayer culture using immunocytochemistry. That of BrdU, PCNA and VEGF was still detected in the microencapsulated SGC-7901 cells on the 7th and 14th day using immnocytochemistry. On the 21st day. the expression of BrdU and PCNA was found at the outside of spheroid, however that of VEGF gene was found within the whole spheroid using immuno-cytochemistry.Conclusion:The cultured model of microencapsulated HGC SGC-7901 was established. which is three-dimension cell growth pattern and whose gene expression, PCNA and VE-GF was stable. It could be the useful platform to research the interaction between the HGC cells and host microenvironment in future.
