Purpose To study the relationship between E-cadherin, proliferating cell nuclear antigen (PCNA), p53 with the occurring, progressing and prognosis of patients with gastric carcinoma (GO.Methods and Materials Using immunohistochemical staining to examine the expression of E-cadherin,PCNA,p53 in archival wax-embedded specimens of 54 GC.Results The positive expression rate (PRE) of E-cadherin in GC was 44. 4%(19/ 54), the PRE of PCNA in GC was 53. 7%(29/54) and p53 was 55. 6%(30/54). The PRE of E-cadherin in GC of invasive serosa was 28. 0%, significantly lower than it in GC of invasive mucosa or submucosa(85. 7%, P<0. 01); the PRE of p53 in GC of invasive serosa (64. 0%) was higher than it in GC of invasive inherent muscle(31. 6%). The PRE of E-cadherin in GC tissue with well differentiated adenocarcinoma (68. 2%) was higher than it with moderately differentiated (25.0%, P<0. 05), and it with poorly differentia ted (28. 6%,P = 0. 017), respective!/. The PRE of E-cadherin in GC with lymph node metastasis (25. 9%) was lower than it without lymph node metastasis (63. 0% ,P<0. 01) ,there was a difference between the PRE of PCNA in GC with lymph node metastasis (77.8%) and without lymph node metastasis (32. 0%, P<0. 01) , the PRE of p53 with lymph node metastasis (76. 0%) was higher than it without lymph node metastasis (40. 7%, P<0. 01). The PRE of E-cadherin in ffl stage GC (7. 7%) was lower than I stage GC (62. 3%, P= 0.001), and H stage GC (58. 8% ,P = 0. 003) , respectively; the PRE of PCNA in I stage GC (98. 8%) was higher than rj stage GC (47. 1 %, P = 0.008). The PRE of E-cadherin of survival time <1 year (23.1%) was lower than it >5 years (100%, P<0. 05); the PRE of PCNA of survival time <1 year (84. 6%) was lower than it 3 years (61. 9%, P = 0. 019), and >5 years (16.7%, P=0. 009), respectively; the PRE of p53 of survival time 1 year (92.3%) was lower than it 3 years (50.0%, P = 0. 011), 5 years (46.2%, P = 0. 014), and>5 years (16.7%, P = 0. 003), respectively. E-cadherin and PCNA ,PCNA and p53 expressions in GC were correlative with each other (P<0. 01). The PRE of E-cadherin(-) ,PCNA( + ) and p53( + ) in GC was 33. 3% , and had relationship with in-vasive depth, metastasis of lymph node, TNM stage and prognosis(P<0. 01).Conclusions There was a significant correlation between E-cadherin expression down-regulation and PCNA expression up-regulation, PCNA expression up-regulation and p53 expression up-regulation in GC; the expression of E-cadherin,PCNA and p53 had relationship with clinicopathologic factors of GC.
