Objective: To analyze the significance of Fas, NF-κB and VEGF-C in the carcinogenesis and progression of invasive ductal carcinoma of breast, to study the experimental foundation of the prognosis evaluation. Methods: Immunohistochemical SP method was used to determine the expression of Fas, NF-κB and VEGF-C in 59 cases of invasive ductal carcinoma of breast, 17 cases of ductal carcinoma in situ and 20 cases of normal breast tissues, the relations between the expression of them and clinicopathological features and survival rate, and the correlation between Fas, NF-κB and VEGF-C was analyzed. Results: (1) The positive expression rates of Fas in invasive ductal carcinoma of breast, ductal carcinoma in situ and normal breast tissues were 50.8%, 64.7% and 85.0% respectively, the positive expression of NF-κB is 62.7%, 41.2 % and 10.0%, the positive expression of VEGF-C is 71.2% respectively, 47.1% and 0% respectively. (2) The positive expression of Fas in carcinoma tissue was closely correlated with tumor diameter, differentiation degree and expression of Her-2(P≤0.05), but not with age, lymph node metastasis, TNM stages and expression of ER or PR, The positive expression of NF-κB was closely correlated with differentiation degree and lymph node metastasis(P<0.05), but not with age, tumor diameter, TNM stages and expression of ER, PR, Her-2. The positive expression of VEGF-C was closely correlated with lymph node metastasis, TNM stages and expression of Her-2(P<0.05), but not with age, tumor diameter, differentiation degree and expression of ER or PR. (3) According to the method of Spearman rank correlation analysis, the positive expression of Fas have statistical negative relevance with the positive expression of NF-κB in breast cancer(P <0.05,r<0). The positive expression of Fas have statistical negative relevance with the positive expression of VEGF-C(P<0.05,r<0). The positive expression of NF-κB have statistical positive relevance with the positive expression of VEGF-C(P<0.05,r>0). (4) According to the method of Kaplan-Meie survival analysis, as for the 5-year survival functions of breast cancer patients, there is statistical relevance between the positive expression group and negative group of Fas and VEGF-C. There is no statistical relevance between the positive expression group and negative group of NF-κB.Conclusion: The positive expression of Fas and NF-κB are important promoting factors in the occurrence and development of breast cancer, Positive expression of VEGF-C have significant relevance with lymph node metastasis of breast cancer(P=0.014,<0.05), which can be a reference for prognosis assessment of patients.
