Research objective:As one of important anabolic hormones, the level of total testosterone in serum could reflect the production and utilization condition of testosterone of athletes, thus it plays an important role in the selection of athletes, assessment of physiology condition and training loads. Especially for athlete assessment of physiological condition, total testosterone level is a common and the most mature indicator for coaches to modify the training strength and for researchers to adapt nutrition supplement and recovery of physical condition. In sports medicine, a large number of studies involving in total testosterone test of athletes use radio immunoassay (RIA) method. RIA is a main method of testing total testosterone for athlete. Magnetism immunoassay (MAIA) is common used to test hormone level in small hospitals. Nowadays some sports team and researchers also try to use it in testing total testosterone for their athletes. This study aimed to use MAIA to test total testosterone of athletes in methodology aspect, and to make sports researchers understand further the application of MAIA.Object and methods:This paper is aim to study comparison between two test methods of serum testosterone for athletes by the literature material law, experimental method, mathematical statistics and logic analysis as the research method. In this article, we do a methodology evaluation to MAIA and RIA, and test the total testosterone level of210athletes using MAIA and RIA, and do a comparative study with difference independent t test and Pearson correlation analysis and Bland-Altman analysis.Results:(1) Methodology evaluation results:The sensitivities of MAIA and RIA are equal. Recycle rate and interclass CV of MAIA is less than RIA. And furthermore the RIA's test range is wider than MAIA.(2)The determination results of serum testosterone:The results of MAIA and RIA tests are inconsistent, readings of RIA are relatively large, and it's variants of readings are bigger as well.(3) T test results:Whether men and women separate statistics, or common statistics, differences between MIAI and RIA have statistically significant (P<0.001).(4) Pearson correlation analysis:MAIA and RIA have a certain correlation with each other (P<0.001).(5) Bland-Altman analysis:Two methods do not have good consistency. RIA could not be replaced by MAIA.Conclusion and suggestion:Both methods MAIA and RIA are sufficient to satisfy the need of tester population in sport community. However these two methods cannot be substituted by either of them, especially avoid horizontal results comparison between these two methods. Because the evaluation standard is mature, RIA method is still the first option of athlete for serum testosterone test.MIAI has its practical application in sport area, but the application has certain limitation. MAIA method should establish its own evaluation standard in application.The best solution for athlete serum testosterone test is:choose one method, select one test staff, build a portal mini-lab, establish serum testosterone reference variants of athlete team, establish personal file of athletes. Completely understand physiology changes of long term training and contest periods. Longitude comparison helps us to have a more accurate and logical judgment on physiology status, hence for better results in contest.
