Studies On ErChen Decoction By Semi-Bionic Enzyme Extraction Mode

Objective:Take ErChen Decoction as an example to probe into the basic mode onextraction of available compound substances in Chinese herbal medicine by semi-bionicenzyme extraction (SBEE).Methods:First,using orthogonal design to optimize SBEE technical conditions forErChen Decoction,with Aurantiamarin,glycyrrhe-tinic acid,etracts(≤1000),polysaccharideand dry extract as comprehensive evaluated indexes.In addition,to optimize the combinedingredients of extraction methods of Erchen Decoction.Next, using homogeneous design tooptimize SBE technical conditions for Erchen Decoction,and using proportional method tooptimize the alcohol concentration for extraction. Next,compare the4kinds of solution ofSBEE method,SBE method,WE method,AE method on compositions and comprehensiveanalysis the results to judge whether SBEE is the best extrac-tion method for ErchenDecoction.Results:The optimized SBEE extraction conditions are the following:First pretreatingherb with2%enzyme Ⅰo f the total weight for12h,pH values of the water for the thriceestraction is2.0,6.5,9.0,added0.5%enzymeⅢ to the first extraction solvent,added1%enzyme Ⅳ to the third extraction solvent,and thrice extraction time is60min,30min,30minand the temperature of extraction is371℃.The optimal combination of this prescriptionis for herb Aurantii nobilis pericarpium,herb Mume fructus,herb Poria,herb Licorice Rootto be decocted in chorus and herb Rhizoma Pinelliae,herb Zingiberis recens to be decoctedrespectively before the decoction is combined finally.The optimized SBE extractionconditions are the following:pH values of the water for thrice extraction is2.0,6.5,8.0,andextraction time is60min,30min,30min;the optimized alcohol concentration for extractingErchen Decoction is30%; the order of comprehensive evaluated values of the4kinds ofextracting solution is YSBEE>YSBE>YWE>YAE。 Conclusion:This study carried out the compare of multicomponent for SBEE solutionwith SBE solution,WE solution and AE solution,all the results show that SBEE solution is thebest.The established semi-bionic enzyme extraction patterns provide a new way of thinkingand method for compound substances and development for New Chinese Medicines.
