Hypoxia-inducible factor-1(HIF-1)which is a DNA binding protein produced under hypoxia condition increase transcription of glycolysis metabolic enzymes combining with hypoxia reacted element, and make the body adapt to the low oxygen environment. Present researches on this gene were mainly in the aspects of human diseases, adaption of the altitude environment and training effectes under hypoxia. The relationship of the function and meat qualities which are influenced by glycolysis has not been reported. Study on the relationship of the HIF-1αgene and the meat qualities could further understand the function of HIF-1αgene, and provide the theory basis for molecular marker assisted selection.209 Duroc×Landrace×York hybrid pigs were randomly selected to slaughter. Meat quality traits, including drip loss, pH value, flesh color, shear force and IMF, were measured by conventional method. The 12th exon of HIF-1αgene of ten species including swine were analyzed by bioinformatics methods using Bioedit, BLAST, ProtParam and NetPhos2.0 Serve softwares and on line tools. SNPs of HIF-1αgene were screened by PCR-SSCP technology. And the relationship of different traits was analysis by relative analysis. The relationship of genotypes and meat quanlity was analyzed by the general linear model. The technology of RT-PCR was used on analysis of the express abundance in shoulder, longissimusdorsi and hind legs. The relationship of meat quality and ratio of relative expression was analyzed by variance analysis and correlational analysis method.The results showed that 130 polymorphic loci were found in the 425bp sequence the 12th exon of HIF-1αgene, and average number of nucleotide differences was maximum in swine spesies. The alanine swiched to threonine because of G-A mutation at 1927 locus, and that may be a new phosphorylation site. 3 SNPs had been screened in parts of HIF-1αgene sequence. The Hardy-Weinberg equilibrium state existed in the gene at the 3 loci. The results demonstrated that no meat quality was influenced with A543T locus (P>0.05), the shear force of mutant type was lower than wild type in G1411T locus (P<0.05) that belonged to the recessive mutation, a G-A transition existed in G1927A locus, and the mutant type was lower than wild type in the pH value (P<0.05), but it was higher than wild type in intramuscular fat (P<0.05), this transversion maked the alanine swich to threonine. Interactions of the 3 mutational loci existed in regulation of shear force, pH value and intramuscular fat. And the relationship existed in different traits. The result of analysis illustrated expression of the HIF-1αgene was found but no significant difference in shoulder, hind legs and longissimusdorsi. A weak positive correlation existed in ratio of relative expression in hind legs and IMF.The results indicate that genetic diversity existes in HIF-1аgene of swine. And the polymorphism of HIF-1αgene is associated with shear force, pH value and intramuscular fat. The allele of 1141 locus can reduce the shear force, and the allele of 1927 locus can reduce the pH value and increase the IMF. Interactions of the 3 mutational loci exist in regulation of pork qualities. The expression of HIF-1αgene existed in difference muscles, it is validated that the gene has a role on IMF at transcriptional level.
