Selenium (Se) is an essential nutrition trace element for human and animal, which plays an irreplaceable role in the physiological functions. By incorporating into selenoproteins, Se plays a great important role in many aspects of biological functions, including the regulation of metabolism; antagonism against poisonous metallic element; clearance of free radical; oxidant defense; delaying aging; improving the immunity and chemopreventive effects etc. Selenoprotein W (SelW) is an important member of selenoprotein family, however, the exact biological function and the mechanism were unclear so far. Gastrointestinal tract was the main organ for chicken to absorb Se. So the injury of gastrointestinal tract can induced Se deficiency, meanwhile, Se deficiency can cause the injury of stomach muscles, stomach, duodenum, small intestine and rectum etc. But the exact mechanism that SelW regulate the inflammation injury of gastrointestinal tract was unknown.1day old chickens were randomly divided into three groups. The chickens in Se deficiency goup were maintained Se deficient granulated diet containing0.033mg/kg Se; in control or high Se group containing0.15or1.5mg/kg Se. Chickens were killed at15,25,35,45and55days old, respectively, and stomach muscles, stomach, duodenum, small intestine, rectum were collected. By combining the clinical symptoms, changes of autopsy and the further investigations of microstructural or Se content of gastrointestinal tract and the mRNA expression of SelW, inflammation related genes TNF-α、iNOS、PGTE、COX-2and NF-κB, we studied the mechanism that Se-deficiency regulate the inflammation injury of gastrointestinal tract, and the effect of different Se levels on the SelW expression in stomach muscles, stomach, duodenum, small intestine, rectum, which further explained the relationship of SelW and inflammation injury. The mains results were showed as follows:1. By the observation of microstructure, autopsy, clinical symptoms and the detection of Se contents, it proved that selenium deficiency could cause the inflammation injury of stomach muscles, stomach, duodenum, small intestine, rectum, manifesting as necrosis, atrophy, which indicated that Se deficiency enteritis model has been set up successfully.2. The Se content or the expression of SelW mRNA in stomach muscles, stomach, duodenum, small intestine and rectum were regulated by the Se level of diet in a dose-and time-dependent manner.3. Selenium deficiency increased the mRNA expression of PTGE、COX-2、TNF-α、iNOS and NF-κB in stomach muscles, stomach, duodenum, small intestine and rectum, which demonstrated that Se involved in the generation or development of inflammation. 4.By analyzing the relativity of the expression of iNOS、TNF-α、NF-kB、PTGE,COX-2and SelW, that presented a negative relativity, which indicated that SelW may play a role in the regulation of inflammation reaction in Se deficiency enteritis.
