Aconite for the sub-root of Ranunculaceae Aconitum, is famous in Sichuan authentic herbs. In the cultivation process of disease have occurred, and seriously affect the yield and quality of aconite. Carry out research on-Aconitum soil microecological, characteristics, and help to reveal the the Aconitum disease mechanism and to explore its prevention and control measures, to promote the development of the aconite industry health. Analysis of health and illness Aconitum rhizosphere soil" Microbial biomass P, soil enzymes, bacteria, fungi and actinomycetes bacteria at different developmental stages, the results are as follows.Chloroform fumigation extraction method for the determination the Jiangyou Aconitum rhizosphere soil microbial biomass phosphorus content, the results show that the soil microbial biomass phosphorus content has no significant differences between health Aconitum rhizosphere soil and sick Aconitum rhizosphere soil; between different monthsthe amount of phosphorus content is also no significant difference. Showed that the content of the soil microbial biomass phosphorus by seasonal changes and the health status of the soil is small.The growing season Aconitum rhizosphere soil invertase, urease, acid alkaline phosphatase were determined. The results show that the different growing seasons, healthy and sick of soil enzyme activity of each significant difference; soil enzyme also has a strong correlation between them (P<0.05).The dilution plate count method for the determination the number of soil bacteria, actinomycetes and fungi,the results showed that the bacteria as dominant bacteria in the soil, significant difference on the different growth stages of its number of microorganisms The DGGE technique to study the diversity of bacterial and fungal communities, the results show that bacterial diversity than the fungi, and the rhizosphere soil of different origin and health status of different microbial taxa.Bacterial sequencing results show that the Aconitum rhizosphere soil microbial bacteria are mainly distributed in four branches three classes five orders four families seven generas. The similarity of the healthy and sick of soil bacterial species higher, minority origin has a unique bacteria such as Pseudomonas aeruginosa, pectin bacilli. Fungal sequencing results show that the Aconitum rhizosphere soil microbial fungi are mainly distributed in one class three orders seven families six generas. And healthy and the sick plant rhizosphere soil fungi in the types of differences, the prevalence of the plant rhizosphere soil there pathogenic type such as Trichothecium roseum, Gibberella fujikuroi, Lecythophora SP.T4A-S5. Fungal and bacterial sequencing results also showed that the DGGE technique than the traditional pure culture microbial community has a unique advantage.Results show that the correlation between the different origin Aconitum soil micro biological indicators:health Aconitum soil urease activity and invertase activity was negatively related to the activity of acid phosphatase and alkaline phosphatase activity was positively correlatedthe number of bacteria and acid phosphatase activity was negatively correlated, and alkaline phosphatase activity kept a positive correlation; sick Aconitum soil urease activity and alkaline phosphatase activity was negatively correlated with the phosphorus content of biomass and alkaline phosphatase activity was positively correlated, the number of bacteria and invertase activity were negatively correlated.Principal component analysis of health and illness Aconitum soil microecology: invertase, urease and acid phosphatase is an important factor affecting the soil micro-of Aconitum; The cumulative contribution of each factor of health and illness Aconitum soil micro characteristics of the formation of the rate differenceThis study were the determination of the Aconitum soil biological indicators and composition characteristics of the various indicators of Aconitum soil micro explore of Aconitum genuineness of the microecological mechanism and conduct Aconitum root exudates amicrobes the microecology between an interpersonal collaborative mechanism of action to lay the foundation in order to reveal their normal growth and disease pathogenesis.
