Objective(1)To observe the accelerative effect of the recombinant human epidermal growth factor used in rabbits’skin wound.(2) To research if there is accelerative effect dosage responds relationship of the recombinant human epidermal growth factor used in the rabbits’skin wound.(3) To research the best dosage range of recombinant human epidermal growth factor used in rabbits’skin wound.MethodsThe dorsal trauma model of rabbits were made.120skin wounds were made on the back of20rabbits. Then the wounds were designed into experiment and control ones. The experiment wounds were subdivided into groups of high dosage rhEGF(90μg/g), subaltern high dosage rhEGF (60μg/g), middle dosage rhEGF(30μg/g),low dosage rhEGF(10μg/g). The control wounds include isotonic saline group and EGF matrix group. The process of wound healing was observed. Mean healing time and healing area rate was calculated. Morever, the dynamic analysis was performed at different times on wound HYP contents.Results(1)The mean healing time of the wounds:The mean time of the rhEGF wound healing groups was(14.7±0.67)d,(13.9±0.92)d,(12.8±0.99)d,(12.5±0.87)d, respectively, the control groups was(15.5±0.85)d and(15.8±0.90)d, The SNK-q test(P<0.05) show the wound healing was accelerated clearly in wounds treated with10μg/g、30μg/g、60μg/g rhEGF, but between60μg/g and90μg/g,there is no difference on healing time(P>0.05).(2)The area of the wounds:On12th day,the area of the the rhEGF groups wounds were (71.26±4.35)%、(76.77±3.48)%、(92.13±2.45)%、(93.17±4.17)%, The SNK-q test(P<0.05) show the area of wound was reduced clearly in wounds treated with10μg/g、30μg/g、60μg/g rhEGF, but between60μg/g and90μg/g,there is no difference on the area of the wounds(P>0.05).(3)The HYP content in the wounds:On12th day, the HYP content in the rhEGF wounds were (3.01±0.54)μg/mg、(4.33±0.30)μg/mg、(5.70±0.55)μg/mg、(5.75±0.65)μg/mg, The SNK-q test(P<0.05) show the HYP content of wounds were increased clearly in wounds treated with10μg/g、30μg/g、60μg/g rhEGF, but between60μg/g and90μg/g,there is no difference on the HYP content(P>0.05). (4) Histologic appearance:In the high dosage rhEGF(90μg/g), subaltern high dosage rhEGF (60μg/g) wounds,the re-epithelialization was faster and new granulation tissue was more than those of the control groups.ConclusionExternal rhEGF can promote incision repair better. In the dosage range from10μg/g to60μg/g, as the dosage increased, the promotion to wounds was better. In the dosage between60μg/g and90μg/g, there is no difference on promotion to wounds.We can conclude that the best dosage range is between30μg/g and60μg/g.
