Objective:To observe the development of chronic inflammation response and urodynamic changes and the expression of SP and NK-1R of L5-S2spinal cord of the rats with chronic nonbacterial prostatitis(CNP).Methods:After the experimental CNP rat model was made,the urodynamical recordings were done on the45th day,on the60th day and on the90th day. Serum,prostates,bladder and spinal cord were based.The levels of TNF-a,IL-1β,IL-2and IL-10in serum and Prostate tissues in the four group rats were measured by ELISA.Under electron and light microscope,histomorphology changes of rat prostates were observed. L5-S2spinal cord were treated with SP or NK1-R floating immunohistochemical stain bvABC.Results:1Prostatic inflammation in the rats of model grops was induced. Different degree of inflammation and lymphocytic infiltration was found in prostate while different levels of cytokines was detected in serum and Prostate tissues.2Comparison with the normal control group,of the45th day model group,micturition volume(MV),maximal micturition volume(Max MV),vesical compliance(VC) and micturition interva(MI) was significantly deceased(P<0.05), post void residual(PVR) and maximal bladder pressure (Max BP) was significantly decreased of the60th day model group(P<0.05),of the90th day model group,MV and MI was significantly deceased while PVR and VC was significantly decreased (P<0.05).3Comparison with the normal control group, of all the model groups, the optical density(OD) of SP and NK-1R ofL5-S2spinal cord was significantly deceased(P<0.05), The OD of SP of L6-S1and NK-1R of L5-S2spinal cord in the90th day model group was significantly decreased(P<0.05),as compared with the other three groups. Comparison with the45d control group,The OD of SP of S1-S2and NK-1R of L5spinal cord in the60th day model group and SP of L5and S2spinal cord in the90th day model group was significantly decreased(P<0.05).Conclusion: Pathological changes were different in the CNP rat models at different stages.The CNP models exist urodynamic changes,but the underlying mechanism need to be further study and to know the correlation factors do significant benefit in guiding the clinical treatment. chronic prostatitis pain could up regulate the expressions of the excitatory transmitter SP and NK-1receptor in the L5-S2spinal cord,which may be associated with the persistence and generalization of prostatitis pain.
