Objective:1Initially to investigae the species distribution, habitat and reservessituation of Xinjiang Aconitum plant.2To detect the determination Xinjiang AconitumPlant Alkaloids and diester alkaloid content, initially certify the resources of the chemicalcomposition.3Adopting TLC fingerprinting method, preliminaryly to carry out thesimilarity comparison, cluster analysis and principal component analysis of Aconitumplants in Xinjiang.4.Initially to select the species of potential medicinal value and similarwith the existing national legal herbs standard Aconitum carmichaelii Debx. and Aconitumkusnezoffii Reichb., which is of widely distributed and abundantly reserved form XinjiangAconitum species.To provide scientific basis for rational using and development ofAconitum species. Methods:1Reviewed literature and specimens, surveyed speciesdistribution, habitat and status of reserves of Xinjiang Aconitum plant.2Taking Aconitineas reference, determined the total alkaloid content in the Xinjiang Aconitum plants by UVspectropHotometers with acid dyes.3Determined aconitine hypaconitine Aconitine in theXinjiang Aconitum plants by HPLC.4Using high performance thin layer silica gel Gplate, taking n-hexane-ethyl acetate-methanol(6.4:3.6:1) for dilute potassium iodide,show color with thin bismuth iodide solution of potassium, got thin layer of fingerprintwith color, to generate common pattern via the chemical fingerprint analysis systemsoftware, and carryed out similarity analysis, cluster analysis and principal componentanalysis. Results:1Initially found five kinds of Xinjiang Aconitum medicinal plants, ofwhich Aconitum leucostomum Worosch. and Aconitum soongaricum Stapf. in Yili wererich, and worthy of further development and utilization.2UV spectropHotometricdetermined the aconitine between the concentration range of0.01176~0.04704mg/mL hada good linear relationship, the regression equation A=16.484C+0.0592,r=0.9992.Average recovery was100.19%, RSD=1.33%. Sample assay results showed that total alkaloid content of the Xinjiang Aconitum species is similar with the existing legal herbsstandard Aconitum carmichaelii Debx. and Aconitum kusnezoffii Reichb., it was worthyof further studying.3By HPLC mesaconitine had a good linear relationship (r=0.9999)between0.0022~0.0864mg/mL, the average recovery was99.69%, RSD=1.04%;aconitine had a good linear relationship (r=0.9999) between0.0201~2.0120mg/mL,average recovery was100.06%, RSD=0.60%; hypaconitine had a good linear relationship(r=0.9999) between0.0021~0.0840mg/mL, the average recovery was99.68%,RSD=1.08%. Sample assay results showed that Xinjiang Aconitum plants containeddiester alkaloids, but the content and ingredients were different between different kinds.4Screened and optimized the conditions for thin layer chromatograpHy, spots were clearand separation was good; similarity analysis showed that Xinjiang Aconitum had somedifferences in chemical composition between different plant species, which can be broadlydivided into three categories. Among which aconitum leucostomum Worosch. andAconitum apetalum(Huth) B. Fedtsch. had high similarity with Aconitum carmichaeliiDebx. and Aconitum kusnezoffii Reichb.. Conclusion:1Xinjiang Aconitum species hadhigh medicinal value, and deserved for further studied.2UV spectropHotometry wassimple, accurate, and can be used for the determination of total alkaloids from XinjiangAconitum plants.3HPLC method was simple, accurate, high sensitivity, can be used indetermination of the diester alkaloids from Xinjiang Aconitum plants.4Establishedthin-layer chromatograpHic fingerprints laid the methodological foundation for the studyof the fingerprint of Xinjiang Aconitum plants.5Initially selected the species of Aconitumleucostomum Worosch. and Aconitum soongaricum Stapf., which were widely distributedand abundantly reserved form Xinjiang Aconitum species, and had potential medicinalvalue and similar with the existed national legal herbs standard, were worthy of furtherbeing studied.
