Effect Of Qingyitang Treatment On The IL-6, IL-10and Intestinal Permeability In Rats With Acute Necrotizing Pancreatitis

Objective:To investigate the effect and mechanism of Qingyitang on inflammation and gut dysfunction in rats with acute necrotizing pancreatitis (ANP) and provide animal experimental evidence for clinical applications.Methods:72healthy male Sprague-Dawley rats (180-220g weight) were randomly divided into3groups with24rats in each group:sham operation group(S group):acute necrotizing pancreatitis group (ANP group); acute necrotizing pancreatitis group trated with Qingyitang (B group). The modle of ANP was induced by injected some of3.8%sodium taurocholate(0.5ml/100g) by the weight of rats beneath the pancreatic capsule to get the model of ANP. The rats of each group were divided into24h group,48h group and72h group. Eternal nutrition was given to rats in each group12hours after operation. Rats in group B were given Qingyitang (2.5ml/100g) by the weight of rats2times every day. Rats in other groups were treated with same volume of normal saline solution by the weight of rats. Rats were sacrificed by abdominal aorta exsanguinations at24th,48th and72nd hour after the operation. The levels of serum amylase were measrured; other blood serum were restored at-80℃, the blood samples were collected to detect the indexes as follows:(1) IL-6level of serum (2) IL-10level of serum; some blood serum for D-lactate.Results:(I) Serum amylase:Compared to group S, the serum amylase level of group ANP and B were up-regulated notably; the level of group ANP peaked at24h and the level of group B peaked at48h, then both of tham down-regulated slowly. Compared to group ANP, the level of B group was significantly lower at72h (P<0.01).(2) IL-6level of serum:Compared to group S, the serum IL-6level of group ANP and B were up-regulated notably; the level of group ANP down-regulated and peaked at72h; the level of group B peaked at24h and down-regulated slowly. Compared to group ANP, the level of group B dowan-regulated notably at72h (P<0.01).(3)IL-10level of serum: Compared to group S, the serum IL-6level of group ANP and B were up-regulated notably and peaked at72h; the level of group B peaked at24h and down-regulated slowly. Compared to group ANP, the level of B group was significantly lower at48h (P<0.05) and72h(P<0.01).(4) Plasma level of D-lactate:Compared to group S, the D-lactate level of group ANP and B were up-regulated notably and peaked at72h; the level of group B was lower than that of group ANP at72h(P<0.01).(5) The results of corrlelative analyses indicated that the serum level of IL-6and intestinal mucosa permeability had significant difference.Conclusion:1. It could be deduced that possible actions of Qingyitang through two points:(1) Depress the expression of IL-6to inhibit the effect of inflammatory reaction.(2) Suppress the expression of IL-10.
