According to the new curriculum plan of the education ministry, Jiangsu Province started to carry out the the high school students’literacy test since2007. Biological compulsory module is the main content of the test. This paper take a series of investigation and exploration to the analysis and measure research of the high school biology literacy test, aiming to provide the beneficial reference basis for the high school biology curriculum teaching, promoting the overall development of students and the teaching level of teachers, which can also effectively supervise the management and quality of high school curriculum.The analysis of the literacy test uses the method of two-dimension——content and cognition, inspecting the consistency of the literacy test and the content standard. The content dimension classification bases on the17topics which is metioned in the curriculum standard. The cognitive dimension is divided into three levels——understanding, understanding and applicatio, on the basis of verbs using in the curriculum standard. By using the same method, the curriculum standard and the literacy test were coded, and then calculate the alignment index. The result shows that the2009literacy test and curriculum standard has statistically significant consistency, while the other four papers and curriculum standards have varying degrees of significant difference. In the content distribution, themes, such as "metabolism","genetic molecular basis","the basic laws of heredity","the regulation of animal life activity","the body’s internal environment and stationarity", occupy high proportions of the content, and the cognition of curriculum standard mainly focus on the level of understanding.The investigation about circumstances of biology teaching and the teachers’ opinion of literacy assessment and investigation shows that high school biology compulsory module teaching progress which also objectively limits teachers to carry out operational experiments, and students have little class to review and consolidate. Teachers’understandings of literacy assessment are generally in line with the idea of assessment which is advocate in the new curriculum and the curriculum standard of biology. But young teachers’understangdings of assessment and the nature of literacy test are not as clearly and deeply as veteran teachers. In addition, teachers are not clear about. Therefore, teachers should constantly improve their professional quality, and effectively develop high school biology compulsory module teaching. Moreover, with the literacy test for Grade2of high school students, problems, such as conceptual confusion, inference and computational difficulties, low capacity of experimental analysis, insufficient capacity of chart analysis, are easily occurred in the test. Considering these problems, teachers can take proper reviewing and training methods to the student’s weakness during the review stage of literacy test.
