Objective:By observing exercise effects on serum Leptin, the hypothalamus Leptin、OB-Rb、 SOCS-3、NPY mRNA expression of high-fat diet rats, we wants to discussion the exercise on the hypothalamus in obese rats SOCS-3mRNA expression and changes in leptin resistance mechanism for exercise to prevent high-fat diet and exercise to lose weight leptin resistance in rats provide an experimental basis.Method:Six-week-old male SD rats with24, were randomly divided into normal control group, high-fat model group, high-fat60-min exercise group. Each group has eight. Exercise group undertake Non-weight-bearing swimming exercise for10weeks exercise. After10weeks, the detection of lipids (TG, TC, HDL-C, LDL-C), serum Leptin, the hypothalamus Leptin、 OB-Rb、SOCS-3、NPY mRNA expression, we wants to discussion the exercise on the hypothalamus in obese rats SOCS-3mRNA expression and changes in leptin resistance mechanism for exercise to prevent high-fat diet and exercise to lose weight leptin resistance in rats provide an experimental basis.Result:1.Weight:Pre-experiment high-fat diet rats in each group body weight was no significant difference.10weeks after the experiment, high-fat model group than the normal control group body weight increased significantly (p<0.05), high-fat60-min exercise group than the high-fat model group body weight reduced significantly (p<0.01).This prompted,60-min exercise can be the prevention of obesity in rats fed with high fat diet.2. Lee’s index:10weeks after the experiment, high-fat model group than the normal control group Lee’s index and body fat increased significantly (p<0.05); high-fat60-min exercise group than the high-fat model group Lee’s index and body fat reduced significantly (p<0.05). This prompted,60-min exercise can be reduction the fat content of obesity in rats fed with high fat diet, can be the prevention of obesity in rats.3.Lipid:10weeks after the experiment, high-fat model group than the normal control group TG、TC、LDL-C increased significantly (p<0.05, p<0.01, p<0.05); high-fat60-min exercise group than the high-fat model group TG、TC reduced significantly (p<0.01, p<0.01). High-fat60-min exercise group than the high-fat model group HDL-C increased significantly (p<0.05). 4.Leptin:10weeks after the experiment, high-fat model group than the normal control group serum leptin increased significantly (p<0.05); high-fat60-min exercise group than the high-fat model group serum leptin reduced significantly(p<0.05).High-fat60-min exercise group serum leptin compared to normal control rats was no significant difference in that high-fat meal can cause levels of serum leptin levels significantly increased, while the movement to correct high-fat diet rats elevated serum leptin and reduced, to improve the high-fat diet rats,"leptin resistance".5. Leptin mRNA expression:10weeks after the experiment, high-fat model group than the normal control group the hypothalamus Leptin mRNA expression reduced, but no significant difference; high-fat60-min exercise group than the high-fat model group the hypothalamus Leptin mRNA expression increased significantly (p<0.05). This prompted,60-min exercise can increase the hypothalamus Leptin mRNA expression of obesity in rats fed with high fat diet.6. OB-Rb mRNA expression:10weeks after the experiment, high-fat model group than the normal control group the hypothalamus OB-Rb mRNA expression reduced significantly (p<0.05); high-fat60-min exercise group than the high-fat model group the hypothalamus OB-Rb mRNA expression increased significantly(p<0.05). This prompted,60-min exercise can increase the hypothalamus OB-Rb mRNA expression of obesity in rats fed with high fat diet.7. SOCS-3mRNA expression:10weeks after the experiment, high-fat model group than the normal control group the hypothalamus SOCS-3mRNA expression increased significantly (p<0.01); high-fat60-min exercise group than the high-fat model group the hypothalamus SOCS-3mRNA expression reduced significantly (p<0.05). This prompted,60-min exercise can reduce the hypothalamus SOCS-3mRNA expression of obesity in rats fed with high fat diet.8. NPY mRNA expression:10weeks after the experiment, high-fat model group than the normal control group the hypothalamus NPY mRNA expression increased significantly (p<0.05); high-fat60-min exercise group than the high-fat model group the hypothalamus NPY mRNA expression reduced significantly (p<0.01). This prompted,60-min exercise can reduce the hypothalamus NPY mRNA expression of obesity in rats fed with high fat diet.Conclusion:1.60-min exercise can be the prevention of obesity in rats fed with high fat diet, reduction the fat content of obesity in rats fed with high fat diet, can be the prevention of obesity in rats. 2.60-min exercise can be prevention high-fat diet induced TC, LDL-C and TG of the increased. This prompted,the exercise for high-fat diet to correct lipid metabolism disorder in rats.3. High-fat diet induced obesity serum leptin levels, suggesting that high fat diet can result leptin resistance in the occurrence. But, high-fat60-min exercise fat rat model of high serum Leptin significantly reduced, suggesting that exercise can prevent occurs leptin resistance of high fat diet rats.4. High-fat60-min exercise group than the high-fat model group the hypothalamus Leptin mRNA、OB-Rb mRNA expression increased significantly,the hypothalamus NPY mRNA expression reduced significantly. This suggesting that, exercise can lower levels of hypothalamic OB-Rb expression increased, binding with high levels of leptin increased in brain, through the JAK/STAT pathway for signal transduction step after the inhibition of hypothalamic NPY, decreased expression of hypothalamic NPY, further regulate appetite. At the same time,60-min exercise can reduce the hypothalamus SOCS-3mRNA expression of obesity in rats fed with high fat diet, and reduce the JAK/STAT signaling pathway inhibition, promotion the cytokines signaling. Suggesting that exercise can prevent occurs leptin resistance of high fat diet rats, prevention of obesity in rats.
