Effect Of Chitosan-Se On Biochemical Indexes, Selenoenzyme Activity And GPx4mRNA Expression Level Of Breeder Rooster

This experiment was conducted to investigate the effects of chitosan-se supplementatio onProduction performance,blood indexes,immune performance,selenium content,oxidation resistance andGPx4mRNA expression level of breeder rooster.One hundred and ninety-six150-day-old healthy Hy-Line Brown breeder rooster were randornlyselected and divided into seven groups with four replicates in each group and seven roosters in eachreplicate.Three levels of selenium from different sources(chitosan-se,SC and sodium selenite,SS)wereadded into the basal diet at0.4,0.8and1.2mg/kg(by selenium content)respectively,but no extra seleniumwas added to the diet in the control group.The experiment lasted for35days.The results showed asfollowing:(1) The ADL of breeder roosters was significantly increased under the0.8mg/kg chitosan-se addedselenium level rather than the control group;the ADFI and F/G in each group was slightly lower than in thecontrol group,but there was no significantly diffirence in the control group (P>0.05).(2) The diet added SC had a great influence on plasma biochemical index and plasma hormone;on21stday,the ALB and AKP content of Group SC(0.8) were significantly higher than that in the controlone(P<0.05),the TC content of Group SC(0.8) and Group SS(0.8) were significantly lower than that in thecontrol one(P<0.05);the GH and IGF-1content of Group SC(0.8) were significantly higher than that in thecontrol one(P<0.05),and T4content was significantly lower than that in the control one(P<0.05).on35thday,the ALB,AKP and GLU content of Group SC(0.8) were significantly higher than that in the controlone(P<0.05);the TC content of Group SC(0.8) and Group SS(0.8) were significantly lower than that in thecontrol one and Group SS(0.4),Group SS(1.2)(P<0.05);the GH content of Group SC(0.4) was significantlyhigher than controlgroup and other groups(P<0.05);the IGF-1and T3content of Group SC(0.8) weresignificantly higher than that in the control one(P<0.05);the ACTH and T4content in each group was weresignificantly lower than that in the control one(P<0.05).(3) Compared with the control group,on21st day,the IgA, IgM content,T lymphocyte conversion rateof Group SC(0.8) and IL-2content of Group SC(0.4) were significantly increased(P<0.05);on35th day, heIgA, IgG content,T lymphocyte conversion rate of Group SC(0.8) and IL-2content of Group SC(0.4) weresignificantly increased(P<0.05);but spleen index had no significantly influence with the controlone(P<0.05).(4) The addition of either SS or SC were significantly increased selenium content in cardiac,liver andspleen plasma of breeder roosters(P<0.05),and with an increase of the supplemental selenium level,theselenium content in body of breeder roosters had increased,selenium retention in SC group was higherthan SS group.(5) Compared with the control group,in cardiac,the GSH-Px,T-SOD,CAT activity of Group SC(0.8) andT-AOC activity of Group SC(0.4) were significantly increased(P<0.05),MDA content was significantlydecreased(P<0.05);in liver,the GSH-Px activity and T-SOD,CAT activity of Group SC(0.8) weresignificantly increased(P<0.05),MDA content was significantly decreased(P<0.05);in spleen,the T-SODactivity of Group SC(0.8) were significantly increased(P<0.05),the MDA content of Group SC(0.4) wassignificantly decreased(P<0.05).(6) The TrxR activity in each group was significantly increased compared with controlone(P<0.05);the IDâ… activity of group SS(0.8) and SC(0.8) were significantly increased compared withcontrol one(P<0.05).The GPx4mRNA expression level was significantly increased compared with controlone(P<0.05);the most efficient added selenium level in their diet was0.8mg/kg SC.Collectively,under the experimental condition, it can be concluded that the diet added SC will increasethe ADL and selenium content,improve blood index and plasma hormone,increase immuneperformance,oxidation resistance and GPx4mRNA expression level of breeder rooster.Among them,theorganic selenium is better than that inorganic one,the most efficient added selenium level in their diet was0.4and0.8mg/kg.
