Salvia miltiorrhiza Bunge is one kind perennial Labiatae Salvia medicinal herb. It contains two kinds'major active chemical constituents: water-soluble phenolic acid and lipo-soluble diterpene anthraquinones which have a good effect on antitumor and on preventing and curing cardiovascular disease (CVD) and so on. Chinese medicine Danshen(Rhizoma of Salvia miltiorrhiza Bunge.) is drawing more and more attentions of scientists to apply it in the treatment of disease. Medical markets'demand for Danshen is also rapidly increasing. Strict quality control and higher quality standard are appealing. Salvia miltiorrhiza Bunge are cultivated in our country widely. The quality of Danshen as one kind Chinese medicine and the valuable active constituents vary strikingly according the cultivated varieties, the climatic and edaphic conditions and so on.This thesis focuses on the differences comparison of active constituents and the genetic differences based on the RAPD analysis in the Salvia miltiorrhiza Bunges. Those Salvia miltiorrhiza Bunges were from different resources and had been cultivated in Yangling in Shaanxi province for two years. In order to roughly illustrate the potential relationship between the major active constituents contents diversity and the genetic diversity.The main contents and results are organized as the following:(1) This research adopted one way which is based on methanol as solvent and ultrasonic extraction to extract both water-soluble and lipo-soluble consitituents of Danshen one time. We established a suitable "one time model" for separation and determination of water-soluble and lipo-soluble constituents of Danshen by HPLC, which simplifies the distillation and separation's procedure and saves more time. The accuracy and reliability of different samples'comparison have been noticeably improved.(2) After the acquisition of five groups (20 each) of Danshen which collected from Laiwu Shandong Province, Bozhou Anhui Province, Beijing, Ruicheng Shanxi Province and Anguo Hebei Province, all Danshen samples were cultivated and managed at Yangling Shaanxi Province for two year. HPLC analysis is done to determine all active constituents'contents of total 29 Danshen samples which are randomLy selected 4 to 7 samples from each group. All 29 cultivars were divided into groups based on the characteristic maps of RAPD and cluster analysis.(3) The results showed that the active constituents'contents in Danshen form Laiwu and Bozhou were slightly higher than those of Danshen from other places, after 2-year-cultivation in Yangling of Shaanxi province, under the same cultivating conditions including soil, water and climate and cultivate management. However, there were no significant differences between different groups.(4) Studies on correlation analysis between major chemical constituents in Salvia miltiorrhiza Bunge from different places showed that, dependencies of different lipo-soluble constituents of Danshen are very significant. But, the relationship between the Dihydrotanshinoneâ… and Tanshinoneâ…¡A were poor. Dependencies of different water-soluble constituents of Danshen are also vary significant except for the correlations between Rosmarinci acid and Caffeic acid. So one constituent such as Tanshinoneâ…¡A's content level can reflect the lipo-soluble constituents'content level; Salvianolic acid B's content level can reflect the water-soluble constituents'contents level.(5) The Hierarchical cluster analysis result based on the Danshen RAPD bands diversity shows that, all of the 29 Danshen samples can been clustered according to the resources. It means that Danshens from the same place show up the similarity genetically, Danshens from different places vary genetically. The Hierarchical cluster analysis results based on Tanshinoneâ…¡A and Salvianolic acid B show that, all the 29 samples can not been cluster into 5 kinds according the Danshens resources. We inferred according the results that, the climate, soil, cultivation and management conditions cannot change the genetic constitute, but those conditions can influence Danshen's major active constituents'content. So Danshens with different resources cannot exert its genetic advantage.
