In this paper,four kinds of materials of Liliaceae from Enshi(Polygonatum sibiricum 、 Polygonatum kingianum 、 Polygonatum odoratum 、 Polygonatum cyrtonema)prefecture were tested.The effective content and biomass of the five year seedlings of four kinds of materials were measured.The selenium tolerance of the four kinds of biennial seedlings was investigated,and the optimum species of the genus Polygonatum in the cultivation of selenium rich culture was optimized.In the pot experiment,the content of selenium,the content of lead,arsenic,cadmium and mercury,the content of the main active active components and the growth of each part were studied under the treatment conditions of different concentrations of exogenous and specific heavy metals.The aim of this study is to explore the concentration of exogenous selenium,which can not only fit the growth but also ensure the low content of heavy metals,and provide a theoretical basis for the selenium enrichment and deep development of the Polygonatum sibiricum.The main conclusions of this paper are as follows:1.The content of polysaccharides and saponins in five years old underground part is higher than that in aerial part,and the difference is significant.Therefore,the underground part is preferentially collected when the material is sufficient.The content of Polygonatum sibiricum,Polygonatum kingianum and Polygonatum cyrtonema in the polysaccharide content was significantly higher than that of Polygonatum odoratum.There was no significant difference in the content of the four kinds of flavonoids in the flavonoid content.The contents of Polygonatum sibiricum and Polygonatum kingianum in saponin were significantly higher than those in the other two materials.In the experiment of exogenous selenium applied to four kinds of materials,Polygonatum sibiricum and Polygonatum odoratum had the highest selenium content in the environment of high concentration of exogenous selenium.Polygonatum sibiricum and Polygonatum kingianum had the largest biomass in the environment of high selenium concentration.Synthetically,Polygonatum sibiricum is the best kind of four materials in thecontent of main active ingredients and selenium tolerance,its five-year biomass is1.46 kg,the content of polysaccharide in the underground tuber is 147.34 mg/g,the content of flavonoids is 52.32 mg/g,the content of saponins is 29.56 mg/g,and the selenium content in the high concentration selenium(100 mg/kg)environment is 7.54mg/kg.2.In the case of exogenous selenium,the optimum range of selenium concentration for Polygonatum sibiricum in biomass,leaf area,leaf number,stem elongation,stem diameter,root number and seedling index was 50~60 mg/kg.As the concentration of exogenous selenium increased,the content of selenium in Polygonatum sibiricum showed a specific curve: the selenium content of Polygonatum sibiricum increased gradually with the increase of the concentration of selenium in a certain concentration,but the content of selenium decreased significantly after the threshold reached the threshold.In the range of exogenous selenium 2.5~70 mg/kg,the selenium content of Polygonatum sibiricum increased with the increase of selenium concentration.In the range of 70~125 mg/kg of exogenous selenium,selenium content of Polygonatum sibiricum decreased with the increase of selenium concentration.The selenium content of Polygonatum sibiricum is the highest when the concentration of exogenous selenium is 70 mg/kg,and the content is 31.2 mg/kg.With the increase of exogenous selenium concentration,the regularity of Pb,As,Hg and Cd uptake by Polygonatum sibiricum is different.When the concentration of exogenous selenium is 30~70 mg/kg,the absorption of Pb is reduced.When the concentration of exogenous selenium is 2.5~60 mg/kg,the absorption of As by Polygonatum sibiricum is reduced.When the concentration of exogenous selenium is90~125 mg/kg,the absorption of Hg is reduced,and the concentration of selenium is70~80 mg/kg,and there is no significant difference between the experimental group and the control group.When the concentration of exogenous selenium was 2.5~30mg/kg and 90~125 mg/kg,the absorption of Cd decreased with the concentration of selenium,and there was no significant difference between the experimental group and the control group in the range of 50~60 mg/kg.From the content of selenium and the content of heavy metals,the best concentration of exogenous selenium can be determined to be 60~70 mg/kg.3.According to the comprehensive test results,the best exogenous selenium concentration of Polygonatum sibiricum is 60~70 mg/kg.Different concentrations of exogenous selenium had significant influence on the accumulation of polysaccharides,saponins and flavonoids and the content of selenium in the polysaccharides,and the underground part and the trend on the ground were basically the same.After exogenous selenium was applied,the content of polysaccharide increased first and then decreased with the increase of selenium concentration,and the maximum value was 169.2 mg/g when the selenium concentration was 60 mg/kg.With the increase of selenium concentration,the content of flavone decreased gradually,but the difference was not significant only when selenium concentration was 2.5~10 mg/kg.After the exogenous selenium was applied,the total content of the saponins was increased and then decreased with the increase of selenium concentration,and the maximum value was 14.01 mg/g when the selenium concentration was 70 mg/kg.Exogenous selenium had a significant influence on the selenium content in the polysaccharides of the Polygonatum sibiricum,showing a tendency to increase first and then decrease.The selenium content in the polysaccharide in the range of selenium concentration 60~70 mg/kg was the highest and was significantly different from the other groups.The scavenging ·OH,DPPH and O2-showed that Polygonatum sibiricum Polysaccharide had strong antioxidant effect.Compared with the control,selenium containing Polygonatum sibiricum Polysaccharide has stronger antioxidant activity,and the selenium content has the same trend with the antioxidant activity.4.According to the comprehensive growth condition,selenium content,heavy metal content and effective ingredient content,the suitable selenium concentration of Polygonatum sibiricum is 60 mg/kg.Under this concentration,the flavonoid content is low and the Hg content is higher than that of the control group.If the test requirement is different,the adjustment can be made according to the specific conditions.
