Enterococcus faecalis and Pediococcus pentosaceus have many important functions and role as probiotics, they can be used in the production of dairy products, food preservative, meat fermentation and production of probiotics. Probiotics has been widely used in livestock and poultry breeding, aquaculture as the advantage of green, natural and efficient. So Enterococcus faecalis and Pediococcus pentosaceus used in the production of probiotics will have broad marketprospects. The reports of Enterococcus faecalis and Pediococcus pentosaceus fermentation process were less, the purpose of this article is to explore the Enterococcus faecalis and Pediococcus pentosaceus fermentation process route, andlay a solid foundation for their large-scale production.In this paper, using the principle of metabolic regulation to optimize Enterococcus faecalis and Pediococcus pentosaceus’ seed medium, seed culture conditions and shake flask fermentation medium, fermentation conditions and10L fermenter fermentation conditions and batch fermentation kinetics. The main research content, method and results are as follows:(1) Use the single factor experiment and orthogonal experimental design tooptimize seed medium and seed culture conditions of Enterococcus faecalis andPediococcus pentosaceus. Obtain the best seed medium and culture conditions, on thebest seed process and the culture conditions, Enterococcus faecalis’ viable countincrease to9.1×10~8CFU/mL; Pediococcus pentosaceus’ viable count increase to8.6×10~9CFU/mL.(2) Using the Response Surface Methodology experiment to optimizethe strain fermentation medium and culture conditions of Enterococcus faecalis andPediococcus pentosaceus. Obtain the most important factors of Enterococcus faecalisand Pediococcus pentosaceus fermentation; First, using the two-level design PlackettBurman test to analysis the main factors of fermentation medium, then range the mainfactors through the steepest ascent method. Finally, through the response surfaceMethodology analysis the optimal level of main factors. Followed using the singlefactor method to optimize the fermentation conditions (pH, temperature, liquidvolume, shaking speed, inoculum size) of Enterococcus faecalis and Pediococcuspentosaceus. Identified strains of Enterococcus faecalis, Pediococcus pentosaceusoptimal fermentation medium and culture conditions, and under the best fermentationmedium and conditions, Enterococcus faecalis’ viable count increase to6.4×10~9CFU/mL; Pediococcus pentosaceus’ viable count increase to9.7×10~9CFU/mL.(3) Using the single factor to optimize the dissolved oxygen, the pH and the stirringspeed of Enterococcus faecalis and Pediococcus pentosaceus’ batch fermentation, and establish their stars batch fermentation cell growth dynamics model, using MATLABsoftware to analyze the experimental data, the results show that the model can be wellfitted Enterococcus faecalis and Pediococcus pentosaceus fermentation process.
