Mechanism Of Zhuyexiaotong Granule Restrained Gouty Arthritis By IL-1β And The Clinical Research

Objectives: This subject investigated the effect and Mechanism ofZhuyexiaotong granule restrained gouty arthritis by IL-1β，observing its clinicalcurative effect and security for supplying the theory evidence of Zhuyexiaotonggranule cure gouty arthritis，and enriched prevention and treatment in TCM of goutyarthritis.Methods:1. Building the MSU synovium of ankle joint to study. Used40Vistarrats，and divided them into4groups randomly （n=10），the normal group，the modelgroup，the Colchicine group and the Zhuyexiaotong granule group. Measured the footswelling of rats and the inflammatory index. Detecting the serum IL-1β content withELISA method and observing the morphological changes of ankle joint byhematoxylin-eosin staining.2. Research the acute toxicity test of mice with themaximum dose method.3. The clinical experiment of this subject selected60cases ofacute gouty arthritis patients. Then divided them into treatment group and controlgroup randomly according to random number table method，each group has30patients. After7days，observed the changes of symptoms，signs，the blood routine，the urine routine，ALT，AST，BUN，Scr，UA，ESR and CRP of patients.Result：1. Zhuyexiaotong granule restrained the foot swelling and theinflammatory index than the model group （P<0.05or P<0.01）.Hematoxylin-eosinstaining：the Zhuyexiaotong granule group showed that the synovium of ankle jointstructure was better than the model group in inflammation. IL-1β changed,too.2. Didnot test the LD50through drenched mice with Zhuyexiaotong granule.In the first day，the maximum dose was249.5g/kg，the equivalent of103.75times clinical dosage of adult.3. The treatment group could improve the scores of symptoms and signs，VAS，UA，ESR and CRP. There were significant difference between two groups（P<0.01）.The clinical experiment showed that those indexes declined（P<0.01），and there wereno obvious abnormalities in the blood routine，the urine routine，ALT，AST，BUNand Scr in two groups during the observation period（P＞0.05）.The treatment groupdepressed UA better than the normal group（P＜0.01）.Conclution：1. The Zhuyexiaotong granule was used for the MSU and it couldbe about IL-1β.2. The Zhuyexiaotong granule was safe to mice and good for short-time using.3. The Zhuyexiaotong granule would cure the gouty arthritis.
