[Objective]This research selected Yiqihuoxue Chinese Herbal Medicine (Astragaluspolysaccharides and Panax notoginsenosides) to intervene the Airway Reconstruction of themice model of asthma.Through observing the influence on the change of asthma mice airwaywall structure,bronchial surrounding inflammatory cells infiltration degree,TGF-β1and Ⅲcollagen type expression,the research explores the blocking effect of Yiqihuoxue ChineseHerbal Medicine on airway reconstruction of asthma mice and the possible workingmechanism,and it compares whether there is difference by using the two medicines or byusing each of them separately,which provides new ideas and experimental basis for theclinical medicine.[Methods]60SPF BALB/C mice have been randomly chosen and divided into6groups.Exceptnormal control group,all of the other groups have established the chronic asthma model byusing ovalbumin(OVA) for3times of the peritoneal sensitized and4weeks of atomizingexcitation,dexamethasone group and drug intervention group(Yiqihuoxue Chinese HerbalMedicined share and used alone,the total three groups) in the first30minutes atomization bygroups were intraperitoneal injection of different drugs.In normal control group,physiologicalsaline is used to replace OVA and medicines.After the materials have obtained,theconventional paraffin embeds lung tissue,and HE gets stained.Determination and computetotal area of bronchial wall,airway wall thickness,with the perimeter of bronchia basementmembrane (Pbm) as the denominator for standardization.Half quantitative assessmentbronchial surrounding inflammatory cells infiltration degree.The localization and expression level (indicated by Absorabnce A) of transformation growth factor-β1(TGF-β1) and Ⅲcollagen type is tested by Immunohistochemistry technique.The extent between differentgroups on the airway inflammation and reconstruction has also been made.[Results]Behavior observation:the sensitized mice experienced fast breathing,sneezing,mouthweeks cyanosis and dysphoria after atomization excitation.Asthma model group is the mostobvious,but drug intervention group is lighter than the model group.Pathological observation: HE stain slicing showed bronchial of asthma model grouphave changed obviously: airway wall thickening,epithelial necrosis fall off,smooth musclelayer and basement membrane layer thickening,there is a great deal of inflammatory cellinfiltration.The pathological changes of dexamethasone group are lesser degree.Panaznotoginseseng group and other drug intervention group performance is in between modelgroup and dexamethasone groups.Normal control group no significant pathologicalchanges.Image analysis showed that the total area of bronchial wall/Pbm,airway wallthickness/Pbm,bronchial surrounding inflammatory cells infiltration degree of asthma modelgroup increased significantly compared with normal group (P <0.01),other drug interventiongroups(group D~F)have significant difference compared with normal group and asthmamodel group (P <0.05).Immunohistochemical detection: TGF-β1mainly expressed in the epithelium on theairway mucosa.Ⅲ collagen type mainly distributed airway mucous membrane lower level andbetween mucosal epithelium cells.Image analysis showed that the absorbance of TGF-β1andⅢ collagen type of asthma model group increased significantly compared with normal group(P <0.01),dexamethasone group and drug intervention group(group C~F)have thesignificant difference compared with normal group and asthma model group (P <0.05).[Conclusion]1.The Astragalus polysaccharides and Panax notoginsenosides can alleviate the airwayinflammation of asthma mice,and the effect of combining these two drugs together is moreobvious than using them separately.2.The Astragalus polysaccharides and Panax notoginsenosides can alleviate the degree of airway reconstruction of asthma mice,and there is no distinction between the usingcombination of drugs and using each of them separately.3.The Astragalus polysaccharides and Panax notoginsenosides can suppress theexpression of TGF-β1and collagen type Ⅲ of asthmatic mice.It hints that two kinds ofmedicines could be used to inhibite the airway reconstruction by reducing TGF-β1expressionand/or Ⅲ collagen type synthesis.
