The Study On The Inhibition Of SGC-7901 Gastric Cancer Metastasis Of The Tonifying Kidney Plus Strengthening Vital Energy Recipe And The Expression Of MMP-3, MMP-9 And CD44v6 In Tumor Tissue In Nude Mice

Objective Build a gastric cancer cell line SGC-7901 nude mice transplantation tumor model, then analysis the concentration of different doses of the tonifying kidney plus strengthening vital energy recipe on SGC-7901 gastric cancer nude mouse transplantation tumor growth,and the influence of gastric cancer of different doses of the tonifying kidney plus strengthening vital energy recipe in nude mouse transplantation tumor tissue about the expression of MMP-3, MMP-9, and CD44v6 protein.Methods Divided the SGC-7901 model randomly into four groups. NS group.5-FU group, TCM low dose group and the high one.Then observe the tumor’s growth and the conditions in general, after the end of treatment, the mice were sacrificed to observe the abdominal cavity tumor metastasis.dissemination in isolated tumor.Calculate the inhibitory rate、detect serum levels of VEGF and bFGF and MMP-3, MMP-9 and CD44v6 protein in the gastric carcinom.Results The tumor formation rate is 100%. The survival rates of TCM groups were higher than control groups.Compared with Nacl group,the tumor weight of 5-FU group and TCM high dose group decrease obviously.And differences has obvious statistical significance (P<0.01). The differences of tumor weight between TCM low dose group and high dose group was statistically significant (P<0.05), however,TCM low dose group and NS group has no statistically significant difference (P>0.05). The inhibitory rates of three experimental group,TCM low dose group, high dose group and 5-FU group,are 14%、35% and 53%.Detection of serum levels of VEGF and bFGF showed that TCM high and low dose bFGF group were lower than that of NS group, and the difference has statistical significance (P<0.01),P<0.05),and difference between the high and low dose group has no statistical significance (P>0.05); The level VEGF content of two TCM groups was lower than that in the NS group, and the difference is statistically significant (P <0.05),however there has no statistical significance between the high and low dose group (P> 0.05).The IHC shows that the expression of MMP-3 in the TCM groups were lower than that in the NS group (P< 0.05), while the expression of CD44v6 and MMP-9 in the TCM groups was not significantly different from that in the NS group. (P all> 0.05).Conclusion The tonifying kidney plus strengthening vital energy recipe can inhibit the metastasis of gastric cancer cells invading and proliferating,and its action may be related to inhibition of the expression of MMP-3 and reduction of the level of bFGF andVEGF.
