Part IThe Relevance Analysis of Evaluation of Liver Cancer after RFA viaUltrasound and Enhanced CTObjective: It is to investigate the relevance of evaluation of the efficacy of RFA(radio frequency ablation) on liver cancer via ultrasound and enhanced CT.Methods: Select the Second Aggiliated Hospital of Dalian Medical Universityfrom January2008to December2012.124patients with complete clinical diagnosis ofliver cancer patients,Primary tumor108,40metastases,atotal of148lesions.Two weeksof the124patients priorto the RFA,RFA treatment,an ultrasound and enhancedCT,analysis the ablation lesions sonographic changes before and after treatment withenhanced CT scan results.Results: Compared with the situation before RFA, there is a positive correlation(p<0.01) of the two methods in evaluating the ablation zone efficacy image performanceaccording to changes of the degree and time of the lesion damages. The result is alsoabout whether ultrasound is superior to CT in evaluation efficacy.Conclusions: Despite ultrasound and CT have their respective advantages,ultrasound excels as the preferred method to evaluate the efficacy of RFA for its simpleoperations, economy, saving time, less trauma and fewer complications. Part IIThe sense of the expression of the caspase-8in rabbit liver VX2tumor-remnant tissues after radio-frequency ablationtreatment AbstractObjective:To discuss the sense of the expression of the cysteine-containingaspartate-specific proteases (caspase-8) in VX2tumor-remnant tissues of rabbits liverafter radio-frequency ablation.Methods: Choose28rabbits for experiment,3rabbit bears VX2tumor cells,2rabbits inoculate in legs for passage,23rabbits inoculate VX2tumor in liver underultrasound guidance as model. The VX2tumor cells were implanted on the inside of thehind legs muscles of rabbits. After two weeks the implantation substantial mass werepalpable, And those rabbits were tumor-bearing rabbits.Cut the fish-like organization atthe size of near1mm3pieces from the edge of tumor in the leg.VX2tumor wereimplanted in23tablets by ultrasonography guidance.Models of rabbit liver VX2tumorwere set up. The random samples with HE staining of two death were observed bymicroscope to determine whether the tumor were inoculated successfully. Models ofrabbit VX2liver tumor were divided into4groups at random. There were3rabbits ingroup A, as the control group with no RFA treatment.Every rabbit in B,C,D,E groupwas subjected to RFA treatment. Tumor-remnant tissues were remained by controllingdamage scope at2/3of the largest radius of the tumor.20rabbits were divided into5groups according to their different time after treatment.That was after RFA group(Bgroup),one week after RFA group (Cgroup), two week after RFA group(Dgroup).There were5rabbits in every group.After observation of tumor-bearingrabbits were executed,four week after RFA group (Egroup).There were5rabbits inevery group.After observation of tumor-bearing rabbits were executed, the expressionstatus of caspase-8in rabbit liver VX2tumor-remnant tissues was detected afterradio-frequency ablation treatment with immuno-histochemical techniques.Results:1.24weeks after inoculation, the ultrasonographic results showed theround or type circular hypoechoic nodules, size1.5+0.5cm, borders clear, no capsule,rich blood flow signals in23rabbit livers. HE staining cells showed nested distribution,large size, irregular, largely and deeply stained nuclei, showing the success ofinoculation in livers of3rabbits chose at random.2. Two hours after treatment, the postoperative RFA ultrasound showed irregular high-level echo, border was not clear and irregular shape at tumor destruction zone.CDF and CDE could not detect blood flow signals.3. Immunohistochemistry results: Before radio-frequency ablation, Caspase-8inVX2tumor-adjacent tissues of rabbit liver was in low level expression with expressionrate9.4±2.3%. Two hours after the radio-frequency ablation, the expression rate ofcaspase-8in tumor-remnants tissues was12.6±2.9%. One week after theradio-frequency ablation, the expression rate of caspase-8in tumor-adjacent tissues was45.8±12.8%. Two weeks after treatment, the expression rate was76.7±15.5%. Fourweeks after treatment, the expression rate was96.6±18.3%. caspase-8of group B-E inthe tumor tissue treated with RFA showed low expression.Conclusions: The radio-frequency ablation can induce apoptosis of liver tumorcells in tumor-adjacent tissues, and thereby can play a more active role in the treatment.
