ObjectivesTo investigate correlation between VDR gene polymorphism and Chinese schizophrenia; insulin and blood lipid levels were compared before and after4weeks of taking risperidone, and study the correlation between VDR gene polymorphism and insulin, blood lipid changes induced by risperidone.Methods1. GenotypingUsing the technology of PCR-LDR to type four locus of VDR genes Fok-I (RS2228570), Cdx-2(RS11568820), Bsm-I (RS1544410), Apa-I (RS7975232).2. Association study between VDR gene polymorphism and Chinese schizophrenia222cases of patients with schizophrenia as case group, and150cases of healthy people as the healthy control group, were collected. VDR genotypes of the case and control group were respectively classified and compared. 3. Correlation between changes of serum lipid and insulin level induced by risperidone and VDR gene polymorphism29cases of first-episode, risperidone monotherapy, hospitalized schizophrenia were collected, fasting insulin and lipid levels were detected before and after taking the medicine for4weeks. And the VDR genes of29cases were typed.All the data were analyzed by SPSS20.0software.Results1.Association between VDR gene polymorphism and Chinese schizophreniaNo significant difference was detected in the distribution of VDR genotypes and allele frequencies between cases and controls; between the case and control group of the same sex, there was no significant difference in the gene distribution; there was no significant difference in the onset age of each genotype; the average onset age of man and woman were respectively21.99±6.05,24.42±7.97, but no significant difference was detected (P=0.121).2. Association between changes of serum lipid and insulin levels induced by risperidone and VDR gene polymorphismThere were significant differences between4weeks and Oweek in insulin and TG levels after taking risperidone (P<0.05). In different Fok-I genotypes, There are significant differences in insulin levels o (P=0.019 for Oweek, P=0.002for4weeks), and insulin changes were significant different (P=0.005), these changes were higher in patients with FF genotype after treatment. In different Apa-I genotypes patients, TG levels were significant different after4weeks risperidone treatment (P=0.030), and there are significant difference in TC and TG changes after treatment, they were larger in patients with AA genotype after risperidone treatment.Conclusion1. There is no correlation between four loci of VDR gene Fok-I, Cdx-2, Bsm-I, Apa-I and Chinese schizophrenia2. The insulin and TG level were elevated significantly after taking risperidone for4weeks3. Fok-I was associated with insulin rise after risperidone treatment, and the insulin increase tend to be higher with the CC genotype; homozygous carriers for the A-allele of the Apa-I SNP tended to be more susceptible to risperidone-induced dyslipidemia.
