| Objective:To monitor the changes of myeloperoxidase(MPO)〠malondialdehyde(MDA) and superoxide dismutase(SOD) levels in the process of myocardial ischemia reperfusion injury(MIRI) in mice.Methods:Twenty male C57BL/6mice were randomized into4group:(1)The sham-operation group;(2)The myocardial ischemia reperfusion group (MIRI group), established by blocking left coronary artery anterior descending branch for30min followed by reperfusion;(3)The leptin pretreatment group (Leptin+MIRI group), established by injecting intraperitoneally leptin at30min before ischemia;(4)The leptin post-treatment group(MIRI+Leptin group); established by injecting intraperitoneally leptin at30min after reperfusion. Blood and heart samples were harvested from each mouse at6hours after reperfusion.We then analyzed the samples for myeloperoxidase(MPO)〠malondialdehyde(MDA)ã€superoxide dismutase(SOD) levels and histological analysis.Results:Administration of leptin before MIRI significantly decreased myeloperoxidase(MPO)ã€malondialdehyde(MDA) levels and averaged up superoxide dismutase(SOD)activity. Conclusions:Leptin pretreatment can reduced the myeloperoxidase (MPO)ã€malondialdehyde(MDA) and superoxide dismutase(SOD) expressions,and significantly reduced tissue death in cardiac mucleã€hemorrhage degree,and reduce the accumulation of the neutrophils. |
