Objective To observe the expression of vascular endothelial growth factor (VEGF) in giant cell tumor of bone (GCT) and to measure the microvessel density (MVD) in tumor tissue, and to investigate the correlation of VEGF expression and MVD with GCT pathology grading (Jaffe grading), surgical staging and prognosis. Methods 34 cases of GCT were enrolled, and the pathology and clinic data were analysed, retrospectively. The VEGF expression and MVD were assessed with immunohistolgy method and the data were performed statistical analysis with the pathology and clinic data. Results VEGF and CD34 were expressed in all cases of GCT investigated;VEGF positive in cytoplasm or cell membrane of mononuclear and multinuclear cells;and CD34 positive in cytoplasm of microvessel endothelial,and the amount of VEGF expression correlated with MVD in GCT (P=0.044); and there was a positive correlation between them (r=0.347); the surgical staging of GCT significantly correlated with the amount of VEGF expression and MVD (P=0.001, P=0.003); The postoperative recurrence rate of GCT correlated with the amount of VEGF expression and MVD (P=0.034, P=0.027); The pathology grading (Jaffe grading), the age of patients , the gender of patients and the position of tumor were not related to the amount of VEGF expression and MVD (P>0.05). Conclusions VEGF expression and MVD are correlated to surgical staging of GCT and the postoperative recurrence rate of GCT, not to pathology grading , not to age , not to gender, and not to the position of tumor. Poor prognosis is indicated by high expression of VEGF and high MVD, which may be the indicators of GCT prognosis.
