Objective: Cervical Cancer is one of the most common and lethalgynecological malignancy, Secondly incidence in female malignant tumors,second only to breast cancer.Its pathological types include squamouscarcinoma,adenocarcinoma,gland scale cancer. Its etiology is associated withthe following factors:1.High risk type HPV persistent infection;2.Sex anddelivery times;3.The synergy of Chlamydia trachomatis,Herpes simplex virusII,Trichomonad infection;4.Smoking,poor nutrition and health conditionsetc.Early cervical cancer is often no obvious symptoms and signs.The cervixmay be smooth or hard to identify with cervical columnar epithelium ectopic.Because of cervical neck patients with normal appearance and easily misseddiagnosis or misdiagnosis.The main transfer channels are direct transfer andlymphatic metastasis with less blood type transfer channels.In cellmetabolism,DNA synthesis and repair and cancer cells rapid appreciation inthe process of splitting folic acid plays an important role,Rapidly dividingcells for folic acid are in high demand,to be used for the synthesis.So in recentyears for transshipment folate in intracellular receptor a has a furtherresearch.Folate receptor binding protein is a kind of film.It in combinationwith physiological levels of folic acid and transport to cells play an importantrole in the process.The expression is lower in normal tissue,highly expressedin tumor cell membrane surface. It divides into FR-α,FR-β,FR-γ, FR-αexpress highly in mucous adenocarcinoma, including ovarian adenocarcinoma,adenocarcinoma of the uterus,cervical carcinoma,testicular choriocarcinomaand certain brain tumors.And rare expression in kidney,colon,and cardiaccancer.About the role of FR-α in the development of cervical cancer and themechanism of action has yet to see related literature reports.Ki-67exists in thenucleus value added,and is closely related to cell proliferation of nucleoprotein. Ki-67as a biomarker for evaluation of cell proliferation,has been widely usedin cancer research and discussion. This experiment uses immunohistochemicalmethod to detect cervical cancer tissues FR-α protein expression and theexpression of ki-67,and discuss the relationship and clinical significance.Methods: Using immunohistochemical method of PV test normalcervical tissue,cervical lesions and cervical cancer tissue before FR-α proteinexpression and the expression of Ki-67.Divided into the experimental group,ofwhich33cases are cervical cancer(cervical cancer phase I in17cases,cervicalCancer phase II in16cases).Control group:30cases of normal cervical tissuesand42cases of cervical intraepithelial neoplasia(CINI15cases, CINII13cases,CINIII14cases).The experimental results to judge:FR-αprotein positiveexpression of judgment:the tan is observed in the cell cytoplasm. Semi-quantitative integral method is used to determine the results.According to thefirst dyeing strength score:no staining of0points,light yellow for one point,tanfor two points,brown for three points.According to the positive cell percentagescore again:randomly selected five high power field of vision,each view cellcount of100,positive cells number grading(0-1%=0,1-10%=1,10-50%=2,50-80%=3,80-100%=4), And For the judgement of product of the tworesults:more than two divided into positive expression(+),less than twodivided into negative expression(-).Results:1Normal cervical tissues,precancerous cervical cancer,cervical cancer tissuesFR-α protein expression.Normal cervical tissues,precancerous cervical cancer,cervical cancertissues FR-α protein expresses in the cytoplasm. FR-α protein expression innormal cervical tissues were positive for6.7%.Cervical precancerous tissueFR-α protein expression positive rate was76.2%.Cervical cancer tissue FR-αprotein expression positive rate was81.7%.The three difference wasstatistically significant.(Table1).Cervical cancer and cervical precancerouslesions was not statistically significant difference in compared group.(P>0.05).Normal tissues group and cervical precancerous lesions was statistically significant. In twogroups,FR-α protein expression is related to HPV infections.HPV infectionpostive and negative HPV infection difference between the two groups havestatistical significance.(P<0.05)4Normal cervical tissues,precancerous cervical cancer tissue,cervical cancertissue expression of Ki-67.Normal cervical tissues,precancerous cervical cancer tissue,cervicalcancer tissue of Ki-67express in the nucleus,tan grains.To see a small amountof Ki-67in normal cervical tissue(Fig.4).Cervical precancerous lesions andcervical cancer tissue expression of Ki-67elevated(Fig.5).Ki-67proteinexspression in normal cervical tissues positive rate was0%.Ki-67expressionin cervical precancerous tissue positive rate was71.4%.Ki-67expressionpositive rate was84.8%in cervical cancer tissue.The three difference wasstatistically significant(Table.5). There are correlations between them.Difference wasstatistically significant(P<0.05).In precancerous cervical cancer tissues, bothwere postively correlated.Correlation coefficient r=0.504,P=0.003. There arecorrelations between them.Difference was statistically significant (P<0.05).Conclusion:1FR-α elevated protein expression is involved in the pathogenesis ofcervical cancer.2FR-α protein expression has nothing to do with age, synergy with HPVinfection.3High expression of Ki-67promote the occurrence of the tumors, nosynergy with HPV infection.4FR-α protein expression in cervical cancer and precancerous tissue hascorrelation with Ki-67, FR-α protein maybe participate in the occurrence anddevelopment of the cell proliferation in cervical cancer.
