Objectve: Investigate the expression of ARF(ADP-ribosylation factor) in gastric cancer tissue and detect its clinical significance.Methods: Select surgical resections that pathologically diagnosed as primary gastric cancer specimens and adjacent tissue specimens of 50 patients from December 2013 to June 2014. Immunohistochemical method was used to detect the different ARF protein expression level in gastric cancer and adjacent tissues and study the relevance of ARF protien expression level in gastric cancer and the clinical pathological parameters of gastric cancer patients. Real-time quantitative PCR(RT-PCR, Real-Time quantitative polymerase chain reaction) was used to detect the different ARF m RNA expression level in gastric cancer tissues and adjacent tissues, and explore the discrepancy relevance in ARF m RNA expression level in gastric cancer tissues and adjacent tissues.Result: ARF protein expression in gastric carcinoma was 72%(36/50), significantly higher than the expression of adjacent normal tissue 10%(5/50)(P<0.05). Gastric carcinoma integrated optical density value(IOD 27707.02±17815.72) and the value of adjacent tissues(IOD12704.16±13932.79) are significantly different. The ARF protein expression has no significant correalation from gender, age, location, tumor size and degree of differentiation(P>0.05), but correlated with the depth of invasion, lymph node metastasis and clinical stage(P<0.05). ARF m RNA expression level in gastric cancer tissues is apparently higher compared with adjacent tissues(P<0.05).Conclusion:(1) The ARF protein and m RNA expression level in gastric cancer tissues were significantly higher than in adjacent tissues. The test of ARF protein and gene might be useful as one of gastric cancer biomarkers.(2) The ARF protein expression level has no direct correlation with gender, age, location, tumor size, but related with gastric serosal invasion, lymph node metastasis and clinical stage. The ARF protein expression level may play an important role in gastric cancer generation and development, therefor is likely to be one of the hallmarks of prognosis in gastric cancer patients.
