Gentiana dahurica Fisch.is a medicinal plant of Gentianaceae.The flower of Gentiana dahurica is a characteristic Mongolian medicine.It is mainly used for clearing heat,detoxifying,relieving cough and expectorating phlegm.It mainly treats lung fever,cough,throat fever,sore throat,poisonous fever and distemper.Previously,the chemical constituents of Gentiana dahurica were systematically studied.It was revealed that the main active constituents were triterpenoids,flavonoids,phenolic acids and iridoid terpenoids.Therefore,the purpose of this paper is to continue the previous work,continue the phytochemical research of Gentiana dahurica,further clarify its pharmacodynamic material basis;after clarifying the active ingredients contained in Gentiana dahurica,study the analytical methods suitable for the quality evaluation and quality control of Gentiana dahurica.The chemical constituents of the flowers of Gentiana dahurica were studied.The structures of these compounds were determined by various spectroscopic methods?mass spectroscopy,1H-NMR,and 13C-NMR?and chemical evidences.Eight compounds were isolated and purified from the flowers of Gentiana dahurica.They were identified as 1-hydroxy-3,7,8-trimethoxyxanthone?1?,28-h ydroxy-11-oxoolean-3-palmitate?2?,gentisic acid?3?,2-furan acid?4?,tricin?5?,3-hydroxy-4-methoxybenzoic acid?6?,?3S?-1,2,3,4-Tetrahydro-?-carboline-3-carboxylic acid?7?,and 5-hydroxymethyl-2-furfuraldehyde?8?.Compounds18 were isolated from the Gentiana dahurica for the first time.To optimize the extraction process of total triterpenes,total flavonoids,total phenolic acids,and total iridoids from Gentiana dauricus,establish methods for the determination and determine the content in 10 samples.The contents of total triterpenes were 24.98-92.77 mg·g-1,total flavonoids were 5.90-11.47 mg·g-1,total phenolic acids 9.54-17.04 mg·g-1,and total iridoids were 67.95-155.16 mg·g-1,respectively.The determination methods were proved to be simple,accurate and reliable,and can be used to evaluate the quality and resource utilization of the flowers of Gentiana dahurica.To establish an HPLC method for simultaneous determination of lutonarin,loganin,saponarin,isoorientin,and isovitexin in flowers of Gentiana dahurica.The analysis was performed on Symmetry?C188 column?4.6 mm×250 mm,5.0?m?with acetonitrile?A?-0.1%aqueous solution of methanoic acid?B?as mobile phase and gradient elution.The flow rate was 1.0 mL·min-1,and the column temperature was 35°C,and the detection wavelengths were 245 and 330 nm.The content ranges of lutonarin,loganin,saponarin,isoorientin,and isovitexin in the 19 batches of flowers of Gentiana dahurica were 1.15-7.71 mg·g-1,0.26-9.23mg·g-1,0.13-0.96 mg·g-1,0.92-7.46 mg·g-1,0.59-1.70 mg·g-1,and there were significant differences of the contents of five constituents.The method is simple,accurate,reproducible,can provid scientific basis for the resource control of flowers of Gentiana dahurica.
