Objective:Using gradual rectal route of administration to model animal’s ulcerative colitis.Observe the effect of the Chinese medicine Kui Chang Ning’s prescription and method on Interleukin-6and Interleukin-10in ill-mice’s serum and colonic. Analyze the chemical effect of Kui Chang Ning to conclude the effectiveness of Kui Chang Ning in treating Ulcerative Colitis.Methods:40SPF-mature mice are randomly selected in four groups:normal group, model group, Kui Chang Ning group (referred iChinese medicine group) and SASP group. Each group consists of10mice. After seven days of normal nursing, all the groups start to use TNBS through gradual rectal route of administration to trigger acute ulcerative colitis. Another injection is carried on15days later using30mg/kg to trigger chronicle Ulcerative Colitis. As the model is ready, from the second day, the treatment continues once a day for a week while recording the mice’s mental health, fur, eating habit, body weight, feces including blood samples in feces (Disease Activity Index, DAI). After the treatment period, mice fast for24hours before taken heart blood sample to test for Interleukin-6and Interleukin-10in the serum and colonic.The mice’s colon and colon membrane are then analyzed based on the recorded data. Results:1.The effect of Kui Chang Ning on mouse’s normal stateAfter a week of rectal injection,Chinese medicine group behaves normal and retains their normal fur. Their eating habit and feces quality greatly improve. Their feces no longer have the sign of mucus. The SASP group also behaves normal and retains their fur. There is not much change observed in eating habit. The feces still contain the liquid materials and mucus. The model group shows tiring and fatigue signs. Their skin is not as healthy as other groups and feces contain liquid. The normal group is on good condition and starts to gain weight.2.Observation on the diseaseBefore the treatment, the colon sample of HE color shows:membrane and the underneath layer of the model group, Chinese medicine group, SASP group congest, swell, erode, ulcerate and form a necrotic part. After the treatment, the congestion and edema symptoms of Chinese medicine group and SASP group disappear. The other symptoms are healed in some parts.3.Effect of Kui Chang Ning on Interleukin-6and Interleukin-10in the serum and colonic of mice with ulcerative colitisCompared to the normally healthy group, the serum and colonic Interleukin-6level of the sample group was significantly higher, and the serum and colonic Interleukin-10level significantly decreased-between the two significantly distinctive groups(P<0.01). The serum and colonic Interleukin-6and Interleukin-10levels of the SASP group were significantly different from those of the sample group(P<0.01). Compared to the sample group, the serum and colonic Interleukin-6level of the Kui Chang Ning group was significantly lower (P<0.01) while the serum and colonic Interleukin-10level was significantly higher (P<0.01). Compared to the SASP group, the serum and colonic Interleukin-6level of the Kui Chang Ning group was lower (P<0.01) while the serum and colonic Interleukin-10level was higher (P<0.01).Conclusion:1.Kui Chang Ning effectively treats the sample mice with ulcerative colitis disease caused by TNBS.2.Kui Chang Ning cures ulcerative colitis more effectively than SASP.3.The possible Ulcerative Colitis treatment mechanisms of Kui Chang Ning are:(1) through increasing the serum and colonic Interleukin-10level, decreasing the serum and colonic Interleukin-6level, recovering the immune function in the sample ulcerative colitis mice group;(2) through antiphlogistic protecting mucous membranes up to the point that the ulcerative colitis ulcers are cured.
