Background:Ischemic stroke is one of the common diseases harmful to human health. It may be the complication of Atherosclerosis, hypertension, coronary heart disease and diabetes, and so on. The inflammatory process involved in cascade of damage after ischemic stroke, which may be the main cause of cerebral ischemia reperfusion injury. Early thrombolytic therapy is an effective therapeutic method, but it will be ineffective in the long-term recovery process.A large number of clinical data and experimental studies have shown that acupuncture treatment has unique functionality and significant effect for ischemic stroke in its acute, post-injury, recovery and rehabilitation period.Objectives:This topic adopts rat models of cerebral ischemia reperfusion made by intravascular occlusion blocking method, to test the dynamic changes of Interleukin-1β (IL-1β) and Intercellular Adhesion Molecule-1(ICAM-1) in6different time intervals of12h,24h,48h,72h,96h,144h after acupuncture intervention by different acupoints compatibility of acupuncture, look for the most effective method of acupuncture treatment of ischemia reperfusion inflammatory response, further compare and observe the variation trend of the Interleukin-1β (IL-1β) and Intercellular Adhesion Molecule-1(ICAM-1) in healthy and affected side of rat’s brain tissue, in order to explore in-depth the mechanisms of acupuncture treatment, and provide some experimental basis for clinical treatment.Methods:1The rats were randomly divided into the bland group and the model control group, with10rats in each group. Do neurological score in modeling rats, observe the cerebral infarction situation using TTC staining method, and observe brain tissue morphological changes of rats using the HE staining method.2The rats were randomly divided into the bland group (10rats) and the model group, successful modeling (based on neurological score) rats were randomly divided into the model control group, acupuncture treatment group1(Baihui Ophryon Compatibility Philtrum-philtra Treatment) and acupuncture treatment group2(Baihui Compatibility involved-side Zusanli acupoint Treatment). There are four groups in total, each group was divided into6time intervals of12h,24h,48h,72h,96h,144h, with10rats in each interval. Take out the brain tissue, frozen sections, and detect the IL-1β, ICAM-1expression between healthy and affected side of tissue by immunohistochemical staining.Results:1Model Evaluation:Observing in HE staining microscope, we found that the ischemic core resulted in infarct staining, reduction of the number of cells, pyknosis of nucleolus, nucleolus becomes smaller, irregular shape; interstitial edema; swelling of endothelial cell; and also a lot of grid-like vacuoles can be see.By TTC staining, we found typical missing staining arisen in ischemic brain tissue, it turned white, matched the middle cerebral artery dominated areas, and it was relatively stable before and after the optic chiasm infarct.2Effect of acupuncturing different acupoints on the IL-1β between healthy and affected brain tissue:After cerebral ischemia-reperfusion injury, IL-1β expression of affected brain tissue in the model group showed a typical bimodal pattern, the first peak reached at12h, and then continued to increase, the second peak reached at48h, after at72h there was a brief drop, and then it continued to show a rising trend; IL-1β expression in the model group healthy brain tissue also showed a typical bimodal pattern, the first peak reached at12h, at48h show a slight drop and then continued to increase, the second peak reached at144h.After treating model rats with Baihui Ophryon Compatibility Philtrum-philtra Treatment and Baihui Compatibility Zusanli Acupoint Treatment, the IL-1β expression levels of affected brain tissue had full downward effect; the first peak of IL-1β of the healthy brain tissue expression increased and the second peak expressed in advance.Comparing Baihui Ophryon Compatibility Philtrum-philtra Treatment with Baihui Compatibility Zusanli Acupoint Treatment in model rats, Baihui Compatibility Zusanli Acupoint Treatment can significantly cut down more IL-1β expression level in model rats affected brain tissue.3Effect of acupuncturing different acupoints on the ICAM-1between healthy and affectted brain tissue:After cerebral ischemia-reperfusion injury, ICAM-1expression of the affected brain tissue in model control group showed a bimodal pattern, the first peak reached at24h, and then decreased rapidly, the second peak reached at72h, and continued to increase; ICAM-1 expression of healthy brain tissue in the model group also showed a typical bimodal pattern, its expression trend was similar to the model control group, the first peak reached at24h, and then continued to increase, the second peak arrived at144h.After treating model rats with Baihui Ophryon Compatibility Philtrum-philtra Treatment and Baihui Compatibility Zusanli Acupoint Treatment, the group given Baihui Ophryon Compatibility Philtrum-philtra Treatment’s ICAM-1expression in the affected brain tissue showed increasing trend before at72h, in the healthy brain tissue showed an increasing trend before at48h; the group given Baihui Compatibility Zusanli Acupoint Treatment’s ICAM-1expression in the affected brain tissue showed an increasing trend before at48h, in the healthy brain tissue also showed an increasing trend before at48h.Comparing Baihui Ophryon Compatibility Philtrum-philtra Treatment with Baihui Compatibility Zusanli Acupoint Treatment in model rats, Baihui Compatibility Zusanli Acupoint Treatment can significantly cut down more IL-1β expression trend in model rats affected brain tissue.4The variation trend comparison of IL-1β, ICAM-1of healthy and affected brain tissuewithin each model group: The expression trend comparison of IL-1β and ICAM-1healthy and affected brain tissue of model rats between the model control group, acupuncture treatment group1and2.IL-1β and ICAM-1expression levels of affected brain tissue in each group were lower than those of healthy brain tissue.Conclusion:1. In bilateral brain tissues of model control group, both IL-1β and ICAM-1expression showed a bimodal pattern, which indicates that IL-1β and ICAM-1is ideal molecular markers observing cerebral ischemia inflammation, it may be used to predicate the effect of inflammatory reaction.2. EA (Electro Acupuncture) group (Baihui Ophryon Compatibility Philtrum-philtra Baihui Compatibility Zusanli) play a regulatory role on stress-damage signal chain networks, which affecting IL-1β and ICAM-1expression of the bilateral brain tissue, suggesting that EA may be adjusted to compensate and reduce ischemia-reperfusion injury by inhibiting the body’s excessive stress, reducing the injury process, and promoting contact between cells and mobilizing healthy brain tissue. 3. Comparing with Baihui Ophryon Compatibility Philtrum-philtra, Baihui Compatibility Zusanli can regulate the body’s inflammatory stress-injury reaction better, and it can reduce the inflammatory reaction and inflammatory injury. When EA intervention is prompted, the distal acupoints effect is better than the partial one.4. Comparing the variation trend of IL-1β, ICAM-1of healthy and affected brain tissue within each model group. IL-1β, ICAM-1expression levels of affected brain tissue in each group were lower than those of the healthy brain. This result suggests that acupuncture participate in the process of cerebral ischemia-reperfusion injury by reducing IL-1β and ICAM-1expression of affected brain tissue, in order to inhibit the inflammatory reaction...
