Purpose:Soft tissue injuries are common and frequently occurring in sports. Both drug therapy and physiotherapy are used treatment methods. Usually, one therapy or alternated two methods are applied to treat soft tissue injuries in traditional way. Based on this situation, We can combine drug and physical factors innovatively compared with treatment effect, providing new idea and method to research in soft tissue injuries in which traditional Chinese Medicine combined with modern physiotherapy is the developing trend. From this tentative idea, Treats is selected as research object in this study because it has some significant efficacy such as diminish inflammation, relieve pain and the amount is the largest in external used Compound Chinese Medicine called New Injury Medicine. As to physical factor, we choose ultrasound which has a better treatment effect to soft tissue injuries. Set the ultrasonic intensity at the same low level (0.1W/cm2), studying the effect of penetration amount of Berberine hydrochloride in Treats cream preparation by different ultrasonic frequency, duration of action and chemical penetration enhancers, finding the suitable ultrasonic parameters for Treats and providing new treatment method for soft tissue injuries and scientific experimental basis for the basic of applied research in Pharmacokinetics in Compound Chinese Medicine Preparation used in Ultrasonic Transdermal Drug Delivery.Methods: Treats cream was made by purifying Berberine hydrochloride in Treats roughly in laboratory which was suit for Ultrasonic Transdermal Drug Delivery. Fixing the ultrasonic intensity at0.1W/cm2, we did the vitro transdermal test separately with adding chemical penetration enhancers or not, selecting different ultrasonic frequency from300KHz-1400KHz and blank control group and drawing accept liquid at each3min,5min,10min,15min and20min which were detected the content of Berberine hydrochloride, calculating the cumulative permeation amount by detected Peak area. Using SPSS software to analysis the influence of three factors above to cumulative permeation amount.Results:1. In this study, the best chromatographic condition for Berberine hydrochloride is that Thermo C18Chromatographic column, the mobile phase consisted by acetonitrile and0.1%phosphate (40:60, pH=4.0, adding0.1g Sodium dodecyl sulfate in each100ml phosphate), Detection wavelength is265nm, Velocity of flow is lml/min, Temperature of column is25℃.2. With no chemical penetration enhancers, low intensity ultrasound has a litter penetration function which is related to ultrasonic frequency; Under adding some certain chemical penetration enhancers, the sequence of ultrasonic frequency is800KHz,900KHz,700KHz,600KHz,500KHz,1000KHz,1100KHz,1400KHz,300KHz,1300KHz,400KHz,1200KHz from high to low of cumulative permeation amount of Berberine hydrochloride.3. Without adding some certain chemical penetration enhancers, the cumulative permeation amount with ultrasound is1.57times than which no ultrasound. Under the situation of adding some certain chemical penetration enhancers, the cumulative permeation amount with ultrasound is0.62times than which no ultrasound. Without ultrasound effected, the cumulative permeation amount with adding some certain chemical penetration enhancers is7.82times than which no chemical penetration enhancers. Having ultrasound, the cumulative permeation amount with adding some certain chemical penetration enhancers is4.5times than which no chemical penetration enhancers.4. Results of data analysis by SPSS software showed that all ultrasonic frequency, duration of action and chemical penetration enhancers affected the dependent variable. The contribution rate showed by Eta spuare is chemical penetration enhancers>ultrasonic frequency>duration of action. 5. As to duration of action factor, cumulative permeation amount of Berberine hydrochloride had very significant difference(P<0.01) in3min and10min,3min and15min,3min and20min,5min and15min,10min and20min with all ultrasonic frequency and adding chemical penetration enhancers or not, it had no difference (P>0.05) with3min and5min,5min and lOmin,10min and15min,15min and20min.6. The highest cumulative permeation amount appeared in the800KHz which had very significant difference compared with others;900KHz is the second;500KHz,600KHz,700KHz,1000KHz and1100KHz which five frequency had no difference (P>0.05) with each other are the third; other frequency are the same level called fourth.Conclusion:1. All ultrasonic frequency can improve the cumulative permeation amount of Berberine hydrochloride,but the extent is different from each ultrasonic frequency,among these frequency,800KHz is the best ultrasonic frequency.2. Under the condition of fixed contact method and low certain ultrasonic intensity, all ultrasonic frequency could improve the cumulative permeation amount of Berberine hydrochloride and800KHz is the best ultrasonic frequency. However, compared with the content of Treats cream, cumulative permeation amount of Berberine hydrochloride was still very low.3. Adding some chemical penetration enhancers in Treats cream, and using Ultrasonic Transdermal Drug Delivery method, cumulative permeation amount of Berberine hydrochloride had very significant improvement in short time, and the best ultrasonic frequency is800KHz.4. Treatment time of ultrasound could take10min as interval.
