Objective: To observe effects of exogenous injection of E2and acute exhaustiveexercise on hormone and protein changes in the rat gonadal axis and thePI3K/Akt signaling pathway in the hippocampus, and to do a comprehensiveanalysis of the impact of acute exhaustive exercise and E2on the exercisecapacity, for exploring central fatigue during exercise produce and recoverymechanisms to provide experimental basis.Methods:48male SD rats were randomly divided into sedentary control group(A, n=12), the exhaustive exercise group (n=12), quiet estradiol group (C, n=12) and Exhausted estradiol group (D, n=12). The rats in C, D groups wereinjected E2(4mg/kg)1hour before exhaustive exercise, rats in B, D two groupswere swimmed to exhaustion, after that, they were killed, blood was collectedusing radioimmunoassay contents of serum T, E2, HCG and GnRH, the bilateralhippocampus were collected for analyzing the protein expression of GPR30,PI3K, AKT, BCL-2and eNOS by Western blotting.Results: the injection of E2was significant decreased (P <0.01) the levels of Tin serum, the levels of T in serum was also reduced by acute exhaustive exercise(P <0.05), however, acute exhaustive impaired the effects of E2on T (P<0.05); the expressions of PI3K, AKT and Bcl-2were decreased significantly byacute exhaustive exercise (P <0.05), but the injection of E2increased PI3K, Akt,Bcl-2and eNOS protein expression (P <0.05). Compared with acute exhaustiveexercise, the effect of E2injection on protein expression was dominant (P <0.05). Conclusions: exogenous Injection E2caused T decreasedin serum, andweakening the feedback capability of HPG axis, thereby made the exercisecapacity reduced. The PI3K/AKT pathway was blocked by acute exhaustiveexercise, and the expression of BCL-2, ENOS was down-regulated. Exogenousinjection of E2can activate the PI3K/Akt signaling pathway, and up-regulate theexpression of of BCL-2, ENOS. The exogenous injections of E2on PI3K/Aktsignaling pathway had no significant activation during acute exercise, evenbeing opposite.
