Surface plasmon resonance (SPR) biosensor is an important direction, which has a high sensitivity, without separation and purification, anti-electromagnetic interference and so on. So SPR technology is widely widespread.A research article angle-modulated surface plasmon resonance spectroscopy.The theoretical and experimental analysis, complete the system optimization improvements, First, the accuracy of the laser power stability greater impact on the measurement results, thus increasing the laser power system measurement channel to correct measurements, Secondly, this paper analyzed and compared triangular prism and semi-cylindrical prism, drawn semi-cylindrical prism is more suitable for the system, Finally, establish a rational theoretical model based on system configuration, simulation optimum chromium film thickness was 3nm, the best gold film thickness is 44nm, finally get tested theoretical relationship between the refractive index and electrical resonance angle medium. I designed and PIN preamplifier module, semi-cylindrical sapphire prism and Lab VIEW software system, and finally set up the system to complete the experiment comprises a laser stability test, dual-channel test system stability, system calibration test and measurement repeatability test.By measuring, dual-channel system stability up to 0.3%, with different molar concentrations of glucose solution for system calibration system derived relationship between the refractive index and the resonance angle, linear fit coefficients up to 0.99972. Finally, glycerol solution repeatable measurements, draw system sensitivity 4 × 10-6RIU.
