The cornea tissue analogue which is cultured by the developing tissue engineering technologyin vitro provides the cornea donor and brings the hope for cornea blind patients. At present themajority of tissue engineering cornea using the way of static culture, which exits some problemsuch as it has irregular shape, poor transparency, and difficult quality between batches etc, due tothe manual operation. The study have found that mechanical stimulation plays an important role inthe field of tissue engineer, biology, morphology and other aspects, so a good mechanicalenvironment has a great influence on the growth of body organs and tissues. In addition, the corneatissue transparency is one of the key factors determines whether the cultivation of tissue engineeringcornea is success, but the assessment criteria of cornea transparency is based on the subjectiveevaluation in the most of time,we can’t obtain the objective evaluation with data.Therefore, this paper develops a dynamic cultured system and a detector for transparency oftissue engineering cornea. The dynamic culture can simulate mechanical environment of the corneain vivo, build an ideal micro-environment for cornea tissue growth; The transparency detector cantest the corneal transparency, provide a more objective evaluation standard:(1) Based on the optical principle and virtual instruments, a new method to measurementtransparency of tissue engineering cornea is determined and a measuring instrument for tissueengineering corneal transparency is designed. The instrument is composed of a computer,DAQ(Data Acquisition) card, sensors for illumination and other devices. By measuring andcalculating the visible light, red laser, PMMA, glass, contact lens and pig’s cornea, the correctionvalue of system error is determined and the value of correction value is0.0094, and the value ofprecision error is0.6-1.0%. It is desirable about the accuracy. The value of transparency detected byred laser has a little difference from value by the normal visible light. However, to the special lightsource, the instrument can judge the superiority and inferiority of the transparency of the tissueengineering cornea. Changing to the different wavelength light source, the transparency of thecornea can be obtained the exact value by the wavelength light source.(2) Developed the dynamic cultured system which is composed of the cornea reactor andcontrol system for tissue engineering cornea. We reasonably build the mechanics environment ofthe biological cultured device based on mechanical analysis by the finite element of ANSYS anddesign the internal structure of corneal biological reactor. The control system can set up theseparameters about the pressure, the rotation of the motor and the flow perfusion. Then, put the cornea seed cells-biodegradable carrier bracket into the cultured chamber placed in sterile CO2cultured box and achieve the dynamic culture. At last, designing the static and dynamic experimentfor acellular porcine cornea stroma, we can find acellular porcine cornea stroma using the dynamiccultured system is more transparency, the surface is more smooth, and the collagen fibers is morethe ordering by using the inverted microscope, the instrument for transparency.In a word, this research about the method and the instrument for the transparency of tissueengineering cornea can obtain the objective data of transparency. It is more scientific andconvenient than the currently used method to detect the cornea transparency and expected tobecome a new standard for the measurement of tissue engineering corneal transparency. Developingthe dynamic cultured system about tissue engineering cornea creates a new way of culturing thecornea tissue in vitro. It lays a good foundation for the clinical application, industrialization andstandardization of tissue engineering cornea.
