Camellia oleifera is a unique woody edible oil tree in China, and known as theworld’s four woody plant material with oil palm, olive and coconut oils. Camellia oilis an pure savageness and edible oil.It a emerging materials of cosmetic and medicineindustry. The high oil content Camellia oleifera “ganshi83-2”were taken as thematerials. Using the paraffin method safranin-fast green double staining Combination,studied were observed during embryonic development. Using frozen section method,Nile Red staining and laser scanning confocal microscopy (LSCM) combine toobserve the formation characteristics and distribution of seed oils cotyledon cellsduring the development of oil bodies, oil body size and oil content of seeds ofdifferent cross-sectional area cotyledon cells, and to explore the relationship betweenseed oil content in the seed cotyledon cells with oil bodies; Studied the seeddevelopment, changes in physical and chemical properties of fruits and indicatorscorrelated with oil content, explore the formation mechanism of the seed lipid. Themain findings are as follows:1. Camellia embryonic developmental stages and the corresponding time periodwere globular embryo (29~33WAF), heart-shaped embryo(33~38WAF),torpedo-shaped embryos(38~41WAF), mature embryos(41~54WAF). Endospermdirectly to the primary endosperm nucleus began to divide, the presence of free nuclei,and the formation of callose wall(1~25WAF), Karyotype endosperm cells fill the sacchamber(25~29WAF), separated by the thin membrane, Cells were polygonal, large,small nucleus and disintegration-shaped were absorbed by embryos(29~41WAF),then finally completely disappeared(41~54WAF). Based seed maturation of solublesugar content, protein content, oil content in different sampling periods showedchanges characteristics of the mature seed maturation process is divided into early,mid and late maturity mature in three stages.2. Establishment and optimization the approach of studies the cotyledon cells ofCamellia of the oil body based on LSCM. Concrete methods: FAA fixed15h, frozen sections, Nile Red staining, PBS buffer rinse, producer, were mounted, LSCMobservation, Which the slice thickness was25μm, Nile Red dye concentration was10μg mL-1, staining time was15min. The average diameter of Camellia oil body was2.52±1.99μm observed by this method.3. The different oil content of oil bodies in cotyledon cells were distributed indifferent Camellia. We found the higher the oil content of Camellia, the deeperstaining of the cell, oil bodies in cells arranged closer, the more the number of oilbodies and the distribution of concentration in the central cell stack, without contactwith the wall, the edge of the oil bodies clearly. The lower oil content more shallowstrain cell staining, intracellular oil bodies or distributed in the cell wall edges, orsparsely dispersed in the cell, the fewer the number of oil bodies and discontinuous.Indicates the number of tea seed oil body cotyledon cells, and seed oil contentbetween the cross-sectional area of the existence of significant positive correlation.Cotyledon cells observed developing oil bodies found, oil formation on the cell edge,with the fat synthesis, accumulation and increased oil content, oil bodies increased,and gradually concentrated in the central cell, arranged more closely.4. The number of the Camellia fruit ventricular is3or4, there are about20ovules. With the development of the fruit, the majority of ovules degenerate, leavingonly1to6eventually develop into mature plump seeds. Camellia fruit duringdevelopment from the olive fruit becomes oval shape. Peel color from blue, greenyellow, and finally become red and yellow. Camellia fruit quality growth exist threepeak period:35~40WAF(mid-June~late July),42~46WAF(early August~earlySeptember),48~53WAF(mid-September~late October).5. The results showed that the oil content of the seeds and fruits of Camelliaoleifera seeds various physical and chemical properties of indicators correlated duringseed development(41~54WAF). Dry seed oil content and fruit quality, fresh seedquality, dry seed quality and wet Jen quality, dry Jen quality, fruit longitudinaldiameter, fruit diameter, fresh seeds out was significant positive correlation betweendry seed rate. Dry seed oil content and moisture content of fresh seeds, kernelmoisture content highly significant negative correlation. The oil content and the soluble sugar content was significantly negatively correlated.
